Objective:Asthma is a chronic inflammatory respiratory disease that seriously threatens human health,and its morbidity and mortality has a tendency to increase year by year.Guben Fangxiao Decoction is the national famous TCM pediatric expert Professor Jiang Yuren's empirical foormula,clinical use for many years,obvious curative effect,which can enhance the children patient's physical fitness,prevent asthma attacks,and improve the children patient's quality of life and long-term remission rate of asthma.In the field of biomedicine,the close correlation of interleukin-17A(IL-17A),nterleukin-10(IL-10)and CXC chemokine ligand 10(CXCL10)with the occurrence of asthma has made clearly,and Guben Fangxiao Decoction of anti-asthma treatment mechanism may be related to this.In this study,we use the animal models of asthma remission to study the effects of Guben Fangxiao Decoction on the expression of IL-17A,IL-10 and CXCL10 in mice with asthma remission,and observe its effect on airway inflammation of asthma remission model mice and mechanism of prevention and treatment of asthma.Methods:By using ovalbumin(OVA)sensitization and respiratory syncytial virus(RSV)intranasal-induced method to establish BALB/c female mice models of asthma in remission,the mice were randomly divided into normal group,model group,low,middle,high dose of Guben Fangxiao Decoction group,montelukast group and dexamethasone group.The general behavioral activities of each group were observed.ELISA was used to detect the protein expression levels of IL-17A,IL-10,and CXCL10 in lung tissue.The Real-time PCR was used to detect the mRNA levels of IL-17A,IL-10 and CXCL10 in lung tissue.Finally,the statistical method was used for analysis and processing.Results:The expression levels of IL-17A,CXCL10 in lung tissues of mice in model group were significantly higher than that of normal group(P<0.01),while the expression of IL-10 was significantly lower than that of normal group(P<0.01).The expression levels of IL-17A,CXCL 10 in lung tissues of mice in low,middle and high dose of Guben Fangxiao Decoction groups,montelukast group and dexamethasone group were lower than that of model group(P<0.05),while the expression IL-10 was higher than that of model group(P<0.05).The correlation analysis showed that IL-10 was negatively correlated with IL-17A and CXCL10 expression in the model group in ELISA test(r =-0.006,r =-0.637,P>0.05),and positively correlated between IL-17A and CXCL10 expression(r = 0.896,P<0.05).Conclusion:1.Asthma in remission airway inflammation may be associated with changes in IL-17A,IL-10 and CXCL10 levels.2.The expression of IL-17A,IL-10 and CXCL10 was regulated by Guben Fangxiao Decoction.3.The mechanism of Guben Fangxiao Decoction in treating asthma may be related with the up-regulation on IL-10 levels and down-regulation on IL-17A,CXCL10 levels,thereby reducing the airway inflammation,so as to achieve the purpose of preventing and treating asthma.
