Objective:Bronchial asthma(abbreviated as asthma)is a common chronic respiratory disease.It is a chronic airway inflammatory disease that involves a variety of inflammatory cells and cellular components.The incidence rate of bronchial asthma is increasing year by year.Chronic airway inflammation occurs in various stages of asthma and is often accompanied by infiltration of eosinophils(EOS).Studies have shown that CCR3 receptors are mainly expressed on the surface of EOS,and act in conjunction with chemokines such as Eotaxin-1(CCL11),Eotaxin-2(CCL24),Eotaxin-3(CCL26),RANTES,and so on.The cells aggregate,activate and release a variety of inflammatory mediators to the site of inflammation,leading to the generation and development of airway inflammation.Guben Fangxiao Yin is the experienced therapy prescripted by Professor Jiang Yuren,a famous pediatric expert in China.It has the effect of reinforcing lung and sconsolidating the superficial and invigorating spleen and reducing phlegm.It has been applied in the treatment of childhood asthma for many years.Clinical trials have proved that this formula can be safely and effectively improved children’s constitution,reduce the number of asthma attacks in children.In this study,a model of asthma remission was established to investigate the possible mechanism of Guben Fangxiao decoction in regulating chronic airway inflammation and mucus secretion in asthmatic mice during remission.Methods:The Balb/c female mice were challenged with ovalbumin(OVA)sensitizition and respiratory syncytial virus(RSV)nasal instillation to establish an asthma remission animal model and randomly divided into the normal group,the model group,the low,the medium,the high dose of Guben Fangxiao Yin group and the montelukast group.Real-time PCR was used to detect the expression of Eotaxin-2,CCR3 and Muc5ac mRNA in lung tissue,and Western Blotting was used to detect the expression of P-STAT6 and CCR3 protein.Results:Compared with those in the normal group,the expression of CCR3,Muc5ac mRNA,and P-STAT6 and CCR3 proteins in the lung tissue increased in the model group(p<0.05).Compared with those in the model group,the levels of CCL24,CCR3,Muc5ac mRNA,and P-STAT6 and CCR3 proteins decreased in the high dose of Guben Fangxiao Yin group and the montelukast group(p<0.05).Conclusion:1.This study found that there was a certain degree of eosinophil infiltration and mucus hypersecretion in lung tissues of mice in the model group,which may be related to the increased expression of CCR3 and Muc5ac mRNA in lung tissue;2.Guben Fangxiao Yin can inhibit the activation of STAT6 protein,affect CCL24/CCR3 binding and Muc5ac expression,suppress eosinophil recruitment and mucus hypersecretion,suggesting the mechanisim by which Guben Fangxiao Yin exerted the effect on asthma.
