The Effect Of Guben Fangxiao Decoction And Astragaluspoly Saccharideon LTD4 And LTR1 Of Asthmatic Model At Remission Stage

Objective:Leukotrienes(LTs) and LTR play a pivotal role in the pathogenesis of acute and chronic asthma as potent inflammatory mediators.Montelukast is a leukotriene receptor antagonist, which the most commonly used with definitive therapeutic targets and pronounced treatment effect.To investigate the effect of GubenFangxiao Decoction (GBFXD) on Cysteinyl leukotriene D4(LTD4) and Cysteinyl leukotrienes receptors 1 (LTR 1) in a murine model of Respiratory syncytial virus (RSV)-and OVA-induced asthma by means of the comparison with astragalus polysaccharides(APS).Methods:Mice were sensitized by intraperitoneal injection with 0.2 mL 0.05% OVA on dl,8except the controls.The preventive group be treated with Guben Fangxiao Decoction (12g/kg,crude drugs) by gavage from the first day.Then mice were exposed to 2.5%OVA in a atomizer for 30 minutes per day until d24, followed by atomization every two days for 2weeks. Then the models were divided to 9 groups to be treated with Guben Fangxiao Decoction (12g、24g、36g/kg), montelukast sodium (2.6mg/kg), astragalus polysaccharide(0.1g、 0.3g、0.6g/kg) and distilled water(20ml/kg) per day for 28 days. The serum were needed to detect the expression of LTD4 and LTR1.The lung be collected for HE stain.Results:Compared with the control group, the expression of LTD4 and LTR1 were significantly higher, in the model group (P<0.01). Compared with the model group, GBFXD high dose group treatment significantly reduced airway jnflammation, and depressed the expression of LTD4 and LTR1 (P<0.05),so did APS. However, the montelukast group did the better performance.Conclusion:There are high expression of LTD4 and LTR1 in remission phase of asthma.The GBFXD and APS can impede lung inflammation analogously.
