Purpose: Through animal experiments observe the therapeutic effect of traditional Chinesemedicine compound herba portulacae lotion on2.4dinitrochlorobenzene (DNCB)-induced chronicdermatit acute eczema model and its possible mechanism.Method:The72clean ICR mice were randomly divided into blank control group, modelgroup, control group and compound herba portulacae lotion group(the maximal、middle、minimal),12mice in each group (male),Mouse acute eczema model was established by overcoating50ul7%2.4dinitrochlorobenzene (DNCB) on mice abdomen, then repeatedly one day. Overcoatedwith10ul1%DNCB on the right inner ear of mouse, after5days of first sensitization. Abovecorresponding drugs were given after24h, twice daily. After the end of treatment seven days, putup all mouse to death, ears quality of each mouse, pathological changes of right ear stained byhematoxylin and eosin (H-E) and the dermis infiltration inflammatory cell (neutrophilicgranulocyte and monocyte) were measured.Result:'Histopathology changes of right ear such as mild hyperkeratosis epidermis, epidermalthickening, the few intracellular edema and mild intercellular edema, dermal infiltrationinflammatory cells (mainly mononuclear cells), dilated dermal blood vessels in control group whichwas consistent with chronic eczema skin pathological changes, indicating the success of themodel.'Ears quality of mice was significantly decreased in compound herba portulacae lotion themaximal、middle、minimal dosage groups, and Pifukang lotion group, difference compared with theblank control group, compound herba portulacae lotion the maximal (P<0.01), have a significantdifference; the other groups (P<0.05), have a difference, indicating compound herba portulacaelotion the maximal、middle、minimal dosage groups, and Pifukang lotion group can alleviate DNCBinduced edema of mouse ear.'Ear quality and thick of mice was significantly decreased in each of compound herbaportulacae lotion the maximal group compared with and Pifukang lotion group and compoundherba portulacae lotion the middle group (P<0.05), indicating compound herba portulacae lotionthe maximal group has better effect in reducing DNCB induced mouse ear edema than Pifukang lotion group and compound herba portulacae lotion the middle group changes.'Ear quality and thick of mice was non-significantly decreased compound herba portulacaelotion the middle and minimal dosage group(P>0.05),showed that the two groups is similar fortreating.'such as epidermal mild thickening, the number of dermal inflammatory cell infiltration weresignificantly reduced in compound herba portulacae lotion the maximal and middle dosage groupsand Pifukang lotion group compared with the control group, compound herba portulacae lotion themaximal (P<0.01), have a significant difference; the other groups (P<0.05), have a difference,showed that compound herba portulacae lotion the maximal and middle dosage groups andPifukang lotion group can inhibit of dermal inflammatory cell infiltration in DNCB inducedcontacting eczema model.'The number of dermal infiltration inflammatory cell were significantly low in compoundherba portulacae lotion the maximal group compared with and Pifukang lotion group andcompound herba portulacae lotion the middle group (P<0.05), indicating that compound herbaportulacae lotion the maximal group have better effect on inhibiting dermal inflammatory cellinfiltration on mice contact eczema model induced by DNCB than Pifukang lotion group andcompound herba portulacae lotion the middle group.'Number of infiltration inflammatory cells and mouse ear quality were low in high and dosesof the compound herba portulacae lotion group than middle and minimal dosage group, hassignificant difference was observed between (P<0.05). But compound herba portulacae lotionthe middle and minimal dosage groups has no significant difference was observed (P>0.05).Conclusion: Compound herba portulacae lotion has a better therapeutic effect on acuteeczema mouse model.
