Association Analysis And Changes Of Serum Levels Of Omentin, CTRP3in Obese Patients With Newly Diagnosed Type2Diabetes Mellitus

Objective:To investigate the circulating levels of omentin, adiponectin and CTRP3in subjects with type2diabetes mellitus (T2DM) and obesity, and explore their correlation with obesity, insulin resistance, glucose and lipid metabolism meanwhile.Methods:100patients with newly diagnosed type2diabetes mellitus (T2DM) and80normal glucose regulation (NGR) controls were divided into normal weight subgroups and overweight or obesity subgroups according to body mass index (BMI)(BMI≥25kg/m^2). The level of fasting serum omentin, adiponectin and CTRP3were measured by ELISA method. Meanwhile blood pressure, fasting plasma glucose (FPG),2h plasma glucose in oral glucose tolerance test (OGTT-2hPG), glycated hemoglobm A1c (HbA1c), total cholesterol (TC), triglycerides (TG), high density lipoprotein-cholesterol (HDL-C), low density lipoprotein-eholesterol (LDL-C), and fasting insulin levels (FINS) were also measured, body mass index and waist hip ratio (WHR) were evaluated and insulin sensitivity was assessed by HOMA-IR.Results:(1) The serum level of omentin in T2DM group was significantly lower than that in NGR group [(15.64±9.64)ng/mL vs (22.9±7.75) ng/mL, P<0.05],indeed the overweight or obesity subjects were significantly lower than that of the normal weight subjects (P<0.05). Pearson correlation analyses showed:the serum omentin was negatively correlated to BMI, WHR, FPG,2hPG, TC, TG, FINS, HOMA-IR and TC/HDL, and positively correlated to sex, HDL, adiponectin. Multiple linear regression showed that gender, HDL, TC/HDL and HOMA-IR were independent variables of omentin. (2) Serum CTRP3level in T2DM subjects were significantly higher when compared to NGR controls [(261.63±73.71) ng/mL vs (332.07±90.53) ng/mL, P<0.05]. Furthermore, the Serum CTRP3levels were significantly higher in either T2DM or obesity subjects compared to normal control subjects (P<0.05). Pearson correlation analyses demonstrated that the Serum CTRP3levels were significantly positively associated with SBP, DBP, BMI, FPG,2hPG, TC, TC/HDC, TG, FINS, HOMA-IR (P<0.05) and negtively correlated with the HDL and adiponectin. When subjected to multiple regression analysis, IR and adiponectin were predictive variables of the CTRP3.(3) The serum level of adiponectin in T2DM group were significantly lower than that in NGR group [(7.28±3.31) ug/mL vs (9.7±4.15) ug/mL, P<0.05], actually the overweight or obesity subjects were significantly lower than the normal weight subjects (P<0.05). Pearson correlation analyses showed. The serum adiponectin was negatively correlated to SBP, BMI, W, WHR, FPG,2hPG, TG, FINS, HOMA-IR, TC/HDL and CTRP3, and positively correlated to HDL and omentin. Multiple linear regression showed that TC/HDL, HOMA-IR and CTRP3were independent variables of adiponectin.Conclusion:(1) Serum levels of omentin, adiponectin in obese patients with newly diagnosed T2DM were significantly decreased, while the CTRP3levels were significantly increased. When it happened in obese patients with type2diabetes,the serum levels of these three adipokines changed further.(2) Serum levels of omentin were significantly positively associated with adiponectin, while the adiponectin and CTRP3were predictive variables of each other, which indicated that the synthetization and secretion of these three adipokines might have some potential relationships.(3) Our data indicated that omentin and CTRP3were correlated with obese indexes, insulin resistance and lipid metabolism in different degrees, and were significantly associated with adiponectin simultaneously, which might directly contribute to the underlying pathophysiology of obese and insulin resistance.
