ObjectiveTo detect the changing of serum visceral adipose tissue-derived serpin protease(vaspin) and omentin levels in the cases with different blood sugar and body weight,making an assay of the relationship among these two kinds of adipokines andglucose and lipid metabolism factors and biochemical indicators.so as to explorewhat role do these adipokines play in the progression of obesity and type2diabetes.MethodsFasting serum vaspin and omentin levels were measured by enzyme-linkedimmuno sorbent assay(ELISA) in22cases of type2diabetes withoverweight/obese.25cases of type2diabetes with normal weight,20cases of normalglucose regulation with overweight/obese and21cases of normal glucose regulationwith normal weight.Meanwhile fasting blood glucose(FBG),fasting plasmainsulin(FINS),hemoglobinA1c (HbA1c),2-hour postprandial blood glucose(PG2h),bloodlipid, blood pressure,height,body weight(BW),waistline,hip circumference,werealso measured,and calculated waist-to-hip ratio(WHR),body mass index(BMI),homeostasis model assessment of insulin resistance index(HOMA-IR):HOMA-IR=FBG×FINS/22.5.Results1. T2DM group serum vaspin concentration(1.39±0.39ng/ml)is significantly higherthan NGR group (1.16±0.5ng/ml)(P=0.017);Obese group (OW/OB) serum vaspinconcentration (1.55±0.41ng/ml) is higher than the normal weight(NW)(1.04±0.36ng/ml)(P<0.01);Between male group and female group,there is significantdifference in the serum level of serum vaspin.male group (1.2±0.49ng/ml)is lowerthan female group (1.42±0.36ng/ml)(P=0.04);By Pearson correlation analysis foundthat serum vaspin concentration is positively correlated with BW、BMI、WHR、HOMA-IR、FPG、PG2h、HbA1c(r=0.501、r=0.753、r=0.841、r=0.258、r=0.22、r=0.253、r=0.227p<0.05or p<0.01).there has no correlation among vaspin andAge、FINS、TG、TC、HDL-C、LDL-C.Multiple linear stepwise regression analysis showed that gender、WHR、HOMA-IR independently associated with serumvaspin.2. Serum omentin levels in T2DM are lower than in NGR(17.965±3.215ug/l vs20.037±4.257ug/l)(P=0.011); Obese group(OW/OB) serum omentinconcentration(15.626±2.298ug/l) is lower than normal weight(NW)(21.948±2.108ug/l)P<0.05);There is significant difference in the serum omentin level between differentgender groups,serum omentin concentration in female group (20.309±3.641ug/l) ishigher than in male group (18.104±3.776ug/l)(P=0.009);By Pearson correlationanalysis found that the levels of serum omentn are significantly and negativelycorrelated with BW、BMI、WHR、HOMA-IR、FPG、PG2h、HbA1c(r=-0.834、r=-0.95、r=-0.859、r=-0.255、r=-0.246、r=-0.275、r=-0.28).there has nocorrelation among omentin and Age、FINS、TG、TC、HDL-C、LDL-C.Multiple linearstepwise regression analysis found that BMI、WHR are serum omentin independentlyassociated factors.Conclusions:1.Significantly increased serum vaspin levels in obese and T2DM patients.Inthe present study,vaspin is independently associated with WHR and HOMA-IR.Theseresults indicate that serum vaspin may be related to glucose and lipid disorder.2.Serum omentin concentration in normal weight group is significantly higherthan in obese group..omentin has remarkable correlations with BMI、WHR, thisconsequence suggest that serum omentin may has closed relationship with thedevelopment of obesity.3.Serum vaspin levels are significantly lower in male than in female.and genderis independent variables of vaspin.for this reason,serum vaspin levels may beaffected by gonadal hormones in the body.
