Background:Since the emergence of the TCT technology and the TBS classification, early diagnosis of cervical cancer and the detection rate of cervical precancerous lesions has a substantial increasing. But because that the TCT diagnosis use a method of morphology and with a certain subjectivity, there is a gap with the pathological diagnosis, especially the ASCUS. Studies have shown that the diseased tissue of Cervical cancer and cervical precancerous lesions both have the over-expression of p16So we try the immunocytochemical staining of p16to cervical cell in order to Investigate the value of p16in Improving the diagnostic accuracy of early cervical lesions and the Feasibility of using p16as a Conventional cytology screening auxiliary indicators.Objective:The feasibility of using p16as a new biomarkers to investigate the early cervical lesions in the TCT specimen and t he clinical value of Screening of high-level lesions in ASCUS and LSILMethods:Using immunohistochemical methods to detect the expression of p16in TCT smears of ASCUS, LSIL, HSIL, the SCC and the NILM which are produced by The TIB-AutoPrep1800fully automated liquid-based cytology producer, All patients underwent cervical biopsy. Basing on the results of the histological diagnosis Compare the p16expression in each group.Results:1. p16expression increased with increasing pathologic grading, and LSIL group and the HSIL group difference was statistically significant.2. Statistically significant difference between the TCT joint p16detection and the TCT detection in the Coincidence rate of Histopathological diagnosis.3. p16expression in ASC-SU and LSIL group have a higher sensitivity, specificity, negative predictive value, positive predictive value.Conclusions:1. The high expression of p16is closely related to the SIL classification, there were significant differences in expression of p16in the NILM1group, the SIL group,SCC group.2. The Coincidence rate of Histopathological diagnosis of early cervical lesions by the TCT joint p16detection is higher than the rate of TCT detection. The TCT smear is a good molecular biology research platform and the combination of TCT and Molecular Biology has an important significance for improvingthe positive detection rate of CIN, and screening for early cervical lesions will provide more objective and more sensitive means of checking. 3. p16in high-level cervical lesions diagnosed ASC-US group has a high negative predictive value and positive predictive value. Besides, with the Advantage of samples simple, easily obtained, patients less painful, the p16detection has a important value of repeat screening for early diagnosis of cervical lesions.
