Objective:To study the expression of COX-2 in different cervix tissues(normal cervix, cervical intraepithelial neoplasia(CIN), cervical carcinoma) by Immunohistochemistry and the relationship with clinical-pathological factors. To detect the expression of COX-2 in advanced cervical squamous carcinoma before and after radiotherapy. Methods:Immunohistochemistry was used to test the expression of COX-2 in normak CIN and cervical carcinoma respectively and the expression of COX-2 in squamous cancer before and after radiotherapy. X2 test and Mann-Whiteney test was used, when.P<0.05,it has statistical significance. Result:1.COX-2 was absent in normal cervix, while the positive express rate in CIN and cervical carcinoma was 56.67%,88.52% respectively. The total difference in three groups was significant (X2=39.01, P=0.000<0.05), the difference in every couple group is significant (P<0.05).2. The positive expression of COX-2 in adenocarcinoma was 91.67%. The positive expression of COX-2 in squamous carcinoma was 87.76%.The expression of COX-2 was significantly increased in adenocarcinoma compared with squamous carcinoma(Z=-3.314, P=0.001<0.05). The expression of COX-2 was related to lymph node metastasis,depth of stromal invasion(P=0.03<0.05, P=0.006<0.05) and has no significance in pathological grading, tumor size and clinical stage(P>0.05).3. The expression of COX-2 after radiotherapy was decreased significantly compared with those before radiotherapy (Z=3.192, P=0.001<0.05). In CR, the expression of COX-2 after radiotherapy was significantly decreased(Z=-2.647,P=0.008<0.01). In PR, the expression of COX-2 was decreased in group after radiotherapy compared with the group before radiotherapy(Z=-1.652, P=0.099>0.05).Compared with PR group, the expression of COX-2 in CR group was decreased significantly (Z=-2.923,P=0.003<0.05). Conclusion:1. The expression of COX-2 in different cervical tissue was significantly. It suggested that COX-2 may be invoved in the develop of cervical cancer.2. Compared with squamous carcinoma, the expression of COX-2 was significantly increased in adenocarcinoma. The expression of COX-2 was significantly related to lymph node metastasis,depth of stromal invasion, and has no ralationship with pathological grading, tumor size and clinical stage. It suggested that the expression of COX-2 may be related to metastasis and invasion in cervical cancer.3.The expression of COX-2 after radiotherapy decreased significantly compared with those before radiotherapy. It suggested that the expression of COX-2 was related to the effects of radiotherapy.
