Background and ObjectiveThis study is designed to evaluate the expression of CDK8、Ki-67、P53in cervical cancer, to explore whether there is differenceof CDK8expression between different grade cervical diseases, toidentify the relevance between expression of the three genes and theclinical factors including pathology, clinical stage and prognosis.Alsowe investigate the expression of CDK8in cervical diseases,todiscuss whether it can predict prognosis. MethodsDetect the expression of CDK8in220paraffin-beddedcervical tissues(40cervical cancer tissues,60CIN high-gradedisease,60CIN low-grade disease,20normal cervical tissues) usingEnvision and super-Envision immunohistochemistry. Also theexpression of Ki-67、P53in40cervical cancer tissues. Usingone-way analysis of variance to explore the, linear correlation,bivariate correlation to discuss the relationship among expressionof CDK8、Ki-67、P53. chi-square test and Fisher exact test to analysisthe relationship between the three genes and the clinical factors.Results1. There is no significant difference between CIN low-grade diseaseand normal cervical tissues(P=0.089). Among other groups CDK8expression enhance with disease grade.(P<0.05)2. Positive correlation was observed in the three gene expressionand HPV infection. There is no relationship between CDK8expression and clinical factors of cervical cancer. A positivecorrelation was observed in ki-67expression and tumor celldifferentiation degree, while none was found with other clinicalfactors. A positive correlation was observed in P53expressionand clinical stage, while none was found with other clinical factors. 3. In cervical cancer tissues, CDK8has positive relationship withP53(P=0.004), ki-67(P=0.001), and also P53, ki-67were positivelycorrelated with each other(P=0.010).4.4. For the CIN low-grade disease, there was no significantdifference between CDK8expression between the healing groupand the persistent group(P>0.05). While for the CIN high-gradedisease, CDK8expression of the persistent group is higher thanthe healing group(P=0.035).Conclusion1. Expression of CDK8increases with disease grade. So weindicate that CDK8protein expression may play an important rolein carcinogenesis and evolution of the cervical cancer, which maybe dependent on cell hyperproliferation.2. Positive relationship was observed between expression CDK8、Ki-67、P53protein and HPV infection. While CDK8has norelationship with clinical factors. The expression of P53protein iscorrelated with FIGO stage. which indicates cell hyperproliferationis an important mechanism during the evolution of the cervicalcancer.The expression of ki-67protein is correlated with tumorcell differentiation degree. 3. CDK8may have an important role in predicting the prognosis ofCIN high-grade disease.
