ObjectiveTo investigate the expression of IL-6 and Stat3 proteins in cervical squmaus cell carcinomas and cervical intraepithelial neoplasia,and the relationship with cliniclopathological parameters,we expect that the study will help us to elucidate the carcinogenetic mechanism of cervical squmaus cell carcinomas and provide theretic foundation for the diagnosis and genic therapy.MethodsFormalin-fixed,paraffin-embedded samples including 44 cases of squamous cell carcinomas (ICC),20 cases of cervical intraepithelial neoplasia(CIN)Ⅰ,10 cases of CINⅡ,20 cases of CINⅢand 15 cases of normal cervical tissues (NCE)were studied.Using Streptavidin-Peroxidase(S-P) immunohisto-chemical technique,the author examined the expression of IL-6 and Stat3 in these samples.The data of this study was analyses usingχ2 test and Spearman rank correlation.Results(1)The rate of IL-6 protein expression in NCE,CIN and ICC were 6.7%,54%,75%,respectively.In CIN and ICC,the positive rates of IL-6 protein expression were significantly higher than that of NCE (P<0.05),and there was significant difference in expression rates of IL-6 between CIN and ICC (P<0.05). In CIN,the IL-6 protein expression of CINⅢwas significantly higher than that of CINⅠ,while CINⅡand CINⅢwere no significant difference in rates of IL-6 expression.In ICC ,there were relationship between IL-6 protein expression and histological grade or lymph node metastases(P<0.05,respectively), but not related with age,clinical stage,muscles invasion and vascular space invasion(P>0.05).(2)The rate of Stat3 protein expression in NCE,CIN and ICC were 13.3%,46%,84.1%, respectively. In CIN and ICC,the positive rates of Stat3 protein were obviously greater than that of NCE (P<0.05),and there was significant difference in expression rates of Stat3 between CIN and ICC (P<0.05).In CIN ,there was no significant difference in rate of expression Stat3 between CINⅠand CINⅡ,and the result was similar in the comparison CINⅡand CINⅢ,while there was significant difference in the expression of Stat3 protein between CINⅢand CINⅠ, and there was significant difference the high expression rates of Stat3 between CINⅠ/Ⅱand CINⅢ(P<0.01).In ICC,there was relationship between Stat3 protein expression and histological grade(P<0.05).There was no difference in expression rates of Stat3 in age,clinical stage ,muscles invasion ,lymph node metastases and vascular involvement(P>0.05).(3)There was positive correlatation between the IL-6 and Stat3(P<0.05).ConclusionsThe results suggest that IL-6 and Stat3 protein overexpression may play an important role in the initiation of cervical cancer.The expression of IL-6 protein was strongly associated with the expression of Stat3 protein.There may be a synergetic action among these genes in"IL-6—gp130—Stat3"pathway and provide genic treatment with indicators .
