The Clinical And Efficacy Study Of Dingxuan Paticles Treating For Vertebral Basilar Insufficiency Wind Phlegm Stasis Resistance Type With Vertigo

Part I.Clinical Research:[Objective] To observe the clinical effects and mechanism of Dingxuan particles,which treat for vertigo caused by wind phlegm stasis resistance and vertebral basilar insufficiency.[Methods] The object of study come from inpatients with vertebrobasilar insufficiency wind phlegm stasis resistance type of vertigo, from the Third Affiliated Hospital of Nanjing University of Taditional Chinese Medicine.There are 65 cases meet the standard, from August 2010 to January 2011. The 65 cases are randomly divided into dingxuan treatment group(34cases)and control group(31cases),the treatment group give Chinese medicine dingxuan particles(include:tianma,zhijiangcan,zhishuizhi,chenpi,fabanxia,chuanxiong,gegen,huangqi,manj ingzi,zhishouwu etal), per os,one dose everyday. The control group are given Nimodipine Tablets, per os,30 mg,3 times a day,course of treatment is two weeks.The main observations are trie changes of TCM symptoms points,the flow velocity of vertebral basilar artery from transcranial Doppler detected and VWF,t-PA%PAI in plasma through treatment.【Results】(1)Dingxuan particles can improve syndromes points of vertigo that caused by wind phlegm stasis resistance,and better than Nimodipine. (2)Dingxuan particles can Obviously improve the flow velocity of vertebral basilar artery, better than Nimodipine. Dingxuan paticles also can improve Pluse Index of vertebral basilar artery, better than Nimodipine. (3) Dingxuan particles can Obviously lower the VWF and PAI degree in plasma,rise the t-PA degree,but undifferentiated with Nimodipine group.[Conclusion] Dingxuan particles can improve symptoms of vertigo patients who are caused by vertebrobasilar insufficiency and wind phlegm stasis resistance type in Traditional Chinese Medicine. Dingxuan particles obviously improve the haemodynamics of Posterior cerebral circulation and Vascular protected.Part II. efficacy study:【Objective】Through the observation of the sedation and anti-convulsion of Dingxuan particles and its affect on hypnosis about pentobarbital sodium,also its role in Anti platelet aggregation on* rat, its antidinic effect on cavia, discuss the drug influence in central nervous system and the mechanism of antidinic.[Methods] The sedation,Anti-convulsion, Hypnosis of pentobarbital sodium all take cleaning ICR mice,give the drug continuouly,then assay the influence of convulsions caused by pentylenetetrazol, hypnosis after pentobarbital sodium,P.i,and the influence in the mice spontaneous activity times after treatment. The Anti platelet aggregation take cleaning SD rats,after the last time treatment,take blood from the carotid artery, observe the influence on platelet aggregation induced by ADP. Antidinic effect take cavia as experimental subject, after the last time treatment,make vertigo model,observe the drug whether has effects on the incubation periods of swing,nystagmus and duration time of vertigo.【Results】(1).The spontaneous activity times of the HD group Obviously lower than normal group.(2).The three groups of Dingxuan particles all can lower the rate of platelet aggregation induced by ADP.(3)This experiment do not discover anti-convulsion and straightforward antidinic effection obviously of the Dingxuan particles.【Conclusion】Dingxuan particles has effection of sedation and Anti platelet aggregation obviously,this research do not discover obviously anti-convulsion and straightforward antidinic effection of the Dingxuan particles.
