The thesis consists three parts:Literature summarize, The theory research of the decoction and syndrome of Ephedra, Aconite and Asarum, The empirical study of the decoction of Ephedra, Aconite and Asarum counteracting bradyarrhythmia.1 Literature summarizeThe thesis summarizes the clinical application of decoction of Ephedra, Aconite and Asarum, and studies the clinical recent developments of decoction of Ephedra, Aconite and Asarum treating various bradyarrhythmias, and summarizes the pharmaco-research of decoction of Ephedra, Aconite and Asarum.2 Decoction and syndrome researchThe decoction of Ephedra, Aconite and Asarum is cold transformation of Shaoyin diseases, because it is too summary, and there is not definite explanation about its pathological mechanism, successive dynasties ministers of medicine had different viewpoints about it , later generations developed the application of decoction of Ephedra, Aconite and Asarum further, and extended the scope of clinical application. Based on previous theory research, systematically study the location of disease, the cause and of disease, pathological mechanism , the main syndrome and its keynote of decoction of Ephedra, Aconite and Asarum, and explain my opinions.3 Empirical study3.1 ObjectiveStudy the decoction of Ephedra, Aconite and Asarum how to counteract bradyarrhythmia. Observe the influence of decoction of Ephedra, Aconite and Asarum on hemocuprein (SOD), malondialdehyde (MDA) and nitrogen monoxidum (NO) . Investigate the mechanism of action of decoction of Ephedra, Aconite and Asarum curing bradyarrhythmia to provide scientific foundation for clinical treatment.3.2 MethodsAdopt inderal (Pro) and acetylcholine(Ach) two rat models ,with the purpose to obverse the decoction of Ephedra, Aconite and Asarum how to counteract bradyarrhythmia caused by Pro and Ach.We deploy malonylurea to observe the effect of decoction of Ephedra, Aconite and Asarum on MDA, deploy xanthinoxidase to observe the effect of decoction of Ephedra, Aconite and Asarum on SOD, deploy nitrate reductase to observe the effect of decoction of Ephedra, Aconite and Asarum on NO. 3.3 ResultsThe decoction of Ephedra, Aconite and Asarum can restraint bradyarrhythmia produced by Ach expressly in the scope of 3.5–14g·kg-1, and the depressant effect of flushing dose is the best . Statistics shows that there are very significant deviation (P<0.01).The flushing dose has an advantage comparing to positive control medicine(P<0.05).The tris-dosage of decoction of Ephedra, Aconite and Asarum also have apparente depressant effect on bradyarrhythmia caused by Pro which isβ-receptor blocking pharmacon((P<0.01), low dose can has good effectiveness. The decoction of Ephedra, Aconite and Asarum can raise activity of SOD and depress content of MDA in the blood serum of two rat models expressly, which show that it can weaken oxygen free radicaland protect cardiac muscle from injury. maybe this is one important pathway repressing bradyarrhythmia. The decoction of Ephedra, Aconite and Asarum can raise content of NO in the blood serum of two rat models expressly, following the increase of dosage, the content of NO and the heart rate of rat is raising.3.4 ConclusionsThe decoction of Ephedra, Aconite and Asarum can restraint bradyarrhythmia produced by Ach and Pro expressly, raise the heart rate of rat. The main path of decoction of Ephedra, Aconite and Asarum counteracting bradyarrhythmia is to restrain vagus and stimulate adrenergic nerve.The decoction of Ephedra, Aconite and Asarum can raise activity of SOD and depress content of MDA,can weaken oxygen free radicaland and protect cardiac muscle from injury , maybe which is one important pathway repressing bradyarrhythmia.Comparing to model group, the decoction of Ephedra, Aconite and Asarum can raise content of NO in the rat blood serum expressly. Indicate that which maybe can stimulate adrenergic nerve, repress vagus nerve handled by NO,thereby adrenergic nerve has major effect on dominating heart. The further research is to study the resource of NO, if NO has a physio-scope, if the raised concentration of NO of treatment group is in the scope of physio- concentration.
