ObjectiveThis study established and selecteda stable and ideal model of allergic rhinitis(AR) with kidney-yang deficiency. Then the different effections of Xiaoqinglong decoction and Mahuangxixinfuzi decoction on two different AR models were compared, while prescription corresponded with the syndrome or not, to explore whether the functional mechanism of Xiaoqinglong decoction and Mahuangxixinfuzi decoction in treating AR was by regulating Th1/Th2cell balance.MethodBased on the guinea pig models of AR sensitized by ovalbumin, glucocorticoid, hydroxyurea or adenine was used to establish the complex models of AR with kidney-yang deficiencyat the same time.The living condition, death and allergy symptoms ofanimals were observed. The concentrations of slgE and histamine in serum were tested by ELISA (enzyme-linked immunosorbent assay) and the morphology changes of kidney and testis tissue were observed by HE(hematoxylin-eosin) staining.The three different models were compared to choose the ideal one. Then two AR models with different syndromewere estabishedand gave Xiaoqinglong decoction orMahuangxixinfuzi decoction to treat. The living condition, death and allergy symptoms ofanimals were observed.The concentrations of IFN-y and IL-4 in serum were tested by ELISA.The IFN-γmRNA,IL-4mRNA,T-betmRNA and GATA-3mRNA expression levels in spleen were tested by real-time PCR.The different effections of Xiaoqinglong decoction on simple sensitized model and complex model with kidney-yang deficiencywere compared,as well as Mahuangxixinfuzi decoction.Results:l.The three different Yang deficiency inducers, glucocorticoid, hydroxyurea and adenine could lead to appear yang deficiency symptoms and signs,lose weightand die.In morphology,comparing with the control group,the effectionof glucocorticoid on kidney and testis tissue was small, while which of hydroxyurea and adenine were great.2.Comparing with the control group, thesimple sensitized model appeared normal living condition and significant allergy symptoms,thecomplex model with kidney-yang deficiency appeared both significant yang deficiency signs and allergy symptoms.In two models, they showed the same changes in the concentration of IFN-y or IL-4 and the expression levels of IFN-ymRNA,IL-4mRNA,T-betmRNA or GATA-3mRNA.The concentration of IFN-γ wasdecreased, IL-4 wasincreased, IFN-y/IL-4 was decreased, IL-2 was normal, IL-10 was increased, and the expression level of IFN-ymRNAwas decreased, IL-4mRNAwas increased, IFN-γmRNA/IL-4mRNAwas decreased, T-betmRNA was decreased, GATA-3mRNA was increased, T-betmRNA/GATA-3mRNA was decreased,. There were significant difference between the two models, and the changes in simple sensitized model were great.3.Both Xiaoqinglong decoction and Mahuangxixinfuzi decoction affected the two AR models.① The effection of Xiaoqinglong decoctionon simple sensitized model included improving allergy symptoms and regulating the concentrations of serum cytokines and the mRNAexpression levesl in spleen.The concentration of IFN-y was increased.The concentration of IL-4 was decreased.The IFN-y/IL-4 was increased.The IFN-ymRNA expression level was increased.The IL-4mRNA expression level was decreased.The IFN-ymRNA/IL-4mRNAwas increased. The concentration of IFN-γ was increased.The concentration of IL-4 was decreased.The IFN-y/IL-4 was increased.The IFN-ymRNA expression level was increased. The IL-4mRNA expression level was decreased.The IFN-ymRNA/IL-4mRNAwas increased.The T-betmRNA expression level was increased.The GATA-3mRNAexpression level was decreased.The T-betmRNA/GATA-3mRNA was increased.Conclusion1.Comparing the three different complex models of allergic rhinitis with kidney-yang deficiency, the model induced by hydroxyurea was more stable and suitable.2.This study estabished two AR models with different syndromes.In living condition and allergy symptoms, simple sensitized model was reflected the syndrome with cold and water retention, complex model with kidney-yang deficiency was reflected the syndrome with cold and kidney-yang deficiency.3.The concentration of serum cytokines and the mRNA expression level in spleen between the two models were significant difference, and the changes on simple sensitized model were great than the complex model with kidney-yang deficiency.It might be the immunologic basic subjects in the different syndromes of AR,and their expression levels related to the kind of syndrome.4.In summerise,both Xiaoqinglong decoction and Mahuangxixinfuzi decoction couldeffect the two AR models, but their effections were differences.In simple sensitized model,the effection of Xiaoqinglong decoctionwas great than Mahuangxixinfuzi decoction.In the complex model with kidney-yang deficiency,the effection of Mahuangxixinfuzi decoctionwas great than Xiaoqinglong decoction. The effection of Xiaoqinglong decoction on simple sensitized model was great,whilethe effection ofMahuangxixinfuzi decoction on the complex model with kidney-yang deficiency was great. It showed that the two decoctions would aim at the specific syndrome of AR. If prescription corresponded with the syndrome,we could obtain a significant effect, but if not, no effect.5 Xiaoqinglong decoction and Mahuangxixinfuzi decoction treated AR by regulating the concentration of IFN-y or IL-4 and the expression levels of IFN-ymRNA, IL-4mRNA, T-betmRNA or GATA-3mRNA, so that reversed the imbalanced Th1/Th2 cell.
