With the rapid development of information technology,E-learning,which is a teaching model not restricted by time and space has become more and more population. E-learning is one of the most important teaching models besides traditional education. Because of there is not a uniform rule of E-learning resources, a lot of different level, different direction and different format E-learning resources on the Internet, seriously hindered the development of E-learning. With the more and more requirements of E-learning services, according to the meaning of domain knowledge in E-learning resources, how to provide the learners with top quality E-learning resources, which is full of Internet; how to manage the excellent course resources scientifically, in order to transplant and reuse them between different learning systems and provide top quality E-learning resources; how to organize E-learning in different standards, according to the educational background , interests and hobbies of the learners; how to guide the learners learning effectively in different requirements and standard, according to the architecture of course knowledge. All these are urgent problems in E-learning research at present.In this paper, it mainly discussed the course learning based on ontology and discussed the following research and implementation based on ontology theory: Research on the architecture of educational resources and the key technology standards of E-learning; Research on the knowledge architecture of < Computer Organization Principle >; Research on ontology , the methods of building ontology and using the ontology editor Protégéto build the course ontology of < Computer Organization Principle >; Research on topic relationship in the course ontology of < Computer Organization Principle >, reasoning topic dependence relationship, parallel relationship and reference relationship to get topic dependence set, parallel set and reference set by Jena development kit. Creating knowledge Learning Road in different level based on topic dependence set, parallel set and reference set. Recommending the educational resources which are corresponding topics on the learning road to learner and guiding the study of course. Finally, it designed and implemented a prototype system of < Computer Organization Principle >course learning based on ontology.The prototype system of < Computer Organization Principle >course learning based on ontology implement the management of course knowledge ontology and reasoning of topic dependence relationship, parallel relationship and reference relationship, with the change of topic and the relationships of topic generate knowledge learning road in different level, it can show the topic knowledge learning law clearly, it is great significance for learner to study course knowledge.
