| Purpose:After acupuncture“Danzhong”, both sides “Zusanli”, both sides“Taichong” on the rat model of hyperplasia of mammary glands. Measure nipple diameter size of each group rats. Observe the pathological morphological change of breast tissue of each group rats. Test the protein expression degree of vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(BFGF) in each group rat breast tissue. analysis the correlation between hyperplasia of mammary gland diseases and angiogenesis factor. Compare the differences and effectiveness of the experimental results between acupuncture group and “Rupixiao”group. analysis and discuss the mechanism of the Acupuncture of“Danzhong”, both sides “Zusanli”, both sides“Taichong” inhibiting excessive hyperplasia of breast glands tissue.Material and method:We choosed 40 female, 12 weeks aged and non-pregnant SD rats,and then randomly divided them into four groups, named normal group, model group,acupuncture group, Rupixiao group(that positive drug control group). except normal group,the other three groups was made model, four groups rats was normally bred.The number of days of making model is 25. after the success of the model made. the three group made model was intervened with treatments, normal group rats was grabbed,once everyday; model group rats was bound and immobilized, 15 min everyday,not was gavaged and acupunctured; for the acupuncture group rats, first comfirmed acupoints: the“Danzhong”, “Zusanli”, “Taichong” according to“experimental acupuncture science”, normally acupunctured “Danzhong”,electro-acupunctured both sides “Zusanli”and both sides“Taichong”, the deph of horizontal needling into “Danzhong” is about 3mm, the deph of perpendicular needling into “Zusanli”is about 3mm, the deph of oblique needling into “Taichong”is about 2mm,EA parameter: disperse-dense wave,frequency 2Hz, intensity was accorded with trembling of needle handle, and stimulation for 15 min, once a day;Rupixiao group was gavaged Rupixiao solution, the standard of Rupixiao to rats is0.3g/kg. experimented as the method that every group rats intervened with treatment above. once everyday,six days everyweek, the time of intervention is 4weeks togetherafter the end of treatment, use vernier caliper to measure the second and the third nipple diameter size of each group rats. take the second breast glands tissue. use HE staining to observe the changes of the pathological morphological of every group rat.use Immuohistochemical staining to test VEGF,BFGF protein expression degree of each group rats.Results:after intervened with treatment for four weeks, begin to observe1.the change of nipple diameter of every group ratscompared with the normal group, the nipple diameter of the model group is obviously larger(P<0.01); compared with the model group, the nipple diameter of the acupuncture group and the Rupixiao group is obviously smaller(P<0.01); compared with the Rupixiao group, the nipple diameter of the acupuncture group is obviously difference(P<0.01), the nipple diameter of the acupuncture group is smaller.2.the pathological morphological change of breast tissue of each group ratscompared with the normal group, in the model group, the shape of the acinus of mammary glands is more regular, there are many excretion in the cavity of the acinus of mammary glands. the cell of the acinus is obviously hyperplastic; compared with the model group, in the acupuncture group,and Rupixiao group, the shape of the acinus of mammary glands is obviously more regular, the excretion in the cavity of the acinus is less,the degree of hyperplasia of the acinus cell is alleviatived;compared with Rupixiao group, in the acupuncture group, the shape of the acinus of mammary glands is relatively regular, the excretion in the cavity of the acinus is relatively less, the degree of hyperplasia of the acinus cell is alleviatived.3. the change of the VEGF protein expression in mammary glands of every group ratscompared with the normal group, the degree of the VEGF protein expression in the model group is significantly stronger(P<0.01); compared with the model group,the degree of the VEGF protein expression in the acupuncture group and the Rupixiao group is significantly lower(P<0.01); compared with the Rupixiao group, the degree of the VEGF protein expression in the acupuncture group is statistical significance,the degree the VEGF protein expression is relatively lower(P<0.05).4.the change of the BFGF protein expression in mammary glands of every group ratscompared with the normal group, the degree of the BFGF protein expression in the model group is significantly stronger(P<0.01); compared with the model group,the degree of the BFGF protein expression in the acupuncture group and the Rupixiao group is significantly lower(P<0.01); compared with the Rupixiao group, the degree of the BFGF protein expression in the acupuncture group is statistical significance,the degree the BFGF protein expression is relatively lower(P<0.01).Conclusion:1.Acupuncture“Danzhong”, both sides “Zusanli”, both sides“Taichong can reduce the wide of nipple diameter of the rat model of hyperplasia of mammary glands and the degree of pathological morphological of tissue. infer to the treatment plan that Acupuncture“Danzhong”, both sides “Zusanli”, both sides“Taichong is probable therapeutic to hyperplasia of mammary glands2.The change of the angiogenesis factors(VEGF, BFGF) in mammary glands of every group rats probable have a relationship to the occurrence and development of hyperplasia of mammary glands.3.the treatment plan that Acupuncture“Danzhong”, both sides “Zusanli”, both sides“Taichong have therapeutic effect on hyperplasia of mammary glands, probably through the regulation of the degree of the angiogenesis factors(VEGF, BFGF) in in mammary glands4.the treatment plan that Acupuncture“Danzhong”, both sides “Zusanli”, both sides“Taichong have probably better therapeutic effect than Rupixiao as oral... |
