Objectives: Constipation is one of the common clinical gastrointestinal symptoms. particularly patients with Cardiovascular diseases and Cerebrovascular diseases, diabetes and after surgery, often aggravate and even exacerbate the condition that due to constipation or difficult defecation。Therefore, finding the drug not only promoting physiological defecation, but also without causing diarrhea, which becomes one of the subjects of clinical research in the majority of medical workers. Zengye chengqi decoction was the famous prescription invented by Wujutong in the Qing dynasty, which was improved on the basis of Cheng qi decoction .was first been seen in Treatise of Febrile Diseases of Wenbing Tiaobian. During the clinical work, my Mentors found that the Zengye chengqi decoction has the efficacy of nourishing yin for relaxing bowels, promoting blood circulation and removing blood stasis, compare with other Chengqi decoction that Zengye chengqi decoction effect are to more alleviate, thus extremely suit able for the patient with cardiovascular and cerebrovascular diseases, diabetes and the surgery which stay in bed. The preliminary clinical observations show that the efficacy is sure, without other side-effects. We tidied the literature about the development of Zengye chengqi decoction in clinical application and found that it has been widely applied in medical. Although it's clinical curative effect has been confirmed by the medical personnel, but the animal experiment study of Zengye chengqi decoction has not been reported. We designed this animal experiments, observing time, shapes, grain ,and wet weight of rabbits'defecation, according to a formula section of the water content of the large intestine and the stools, measuring the level of MTL ,to investigate the mechanism of defecation on Zengye chengqi decoction. A preliminary research of Security on electrolytes, hemorheology , so as to provide reliable experiment data to clinical application.Methods: 24 health rabbits, weighing 2.0±0.3kg, both male and female (the female was nonpregnant). the quantitative amount of water and food was provided in the normal daily during a week , from 8:00 to 18:00 2 days consecutively ,the measurement of the total grain and wet weight of defecation , the grain and weight of first defecation in rabbits , had the average of two days as baseline. The 10 day, we divided these rabbits into 3 groups at random, n = 8, A, Zengye chengqi decoction group; B, Forlax(Macrogol) group; C, normal saline group. The study was started at about 8:00 hours ,at 8:00and 12:00intragastric administration .After intragastric administration recorded the time , shape, grain and weight of first defecation in rabbit , collected stool from 8:00 to 18:00 at any time, weighing, The total stools were dried using a drying oven at 90℃for 8 h and their dry weight was measured. The water content of the stools was estimated by comparing the wet and dry weights of the stools. Before and after treatment each blood sample, measured blood rheology, electrolytes and MTL. Before and after treatment, between different groups would be compared. All the animals in the experimental 18:00 with 25% urethane ear vein anesthesia, Open the abdominal cavity, observed residual stool and intestinal peristalsis. Take jejunum, ileum, colon and rectum specimens, remove the contents, weighed, were dried using a drying oven at 90℃for 8 h and their dry weight was measured, according to a formula section of the water content of the large intestine, compared the difference between groups.Results:1 Effect of the stool form of Rabbits: Group A, the first stool maintains the normal shape after intragastric administration, the surface is moist, the texture is softer, stool inside is moist. The time appearance of first bar stool is 5.39±0.20 hours. Group B, the first stool maintains the normal shape after intragastric administration, the surface is moist. The first bar stool happens was 7.48±0.16 hours. Group C, the first stool maintains the normal shape after intragastric administration, the surface is moist, no bar stool happen. Diarrhea muddy stools or watery stools are not observed in three groups.2 Effect of the time of first bar stool in rabbits: After intragastric administration. bar stool can be observed in group A and B ,The stool form isn't change within 10 hours in group C .Compare group A with group B, the difference is significant(p<0.01).3 Observation on residues stool in large intestine in rabbits: Open the abdominal cavity at 18:00 on the day, the rectum has stool accidently in group A. there is a large number of soft stool in the rectum in group B, there is granular dry stool in the rectum in group C.4 Effect of peristalsis of rabbits: after intragastric administration, Compare group A with group B and C, the difference is significant (p<0.01).compare group B with group C, the difference is not significant(P>0.05).5 Effect of time of first defecation in rabbits: After intragastric administration, group A, the difference is significant before and after treatment (p<0.01), group B and C, The difference is not significant before and after treatment (P>0.05).Compare group A with group B and C, the difference is significant (p<0.01), compare group B with group C, the difference is not significant (P>0.05).6 Effect of the weight and grain of first defecation in rabbits: it can increase the weight and grain of first defecation after intragastric administration in each group, and the difference is significant(p<0.05), Compare group A with group B and C, the difference is significant(p<0.01).compare group B with group C, the difference is not significant(P>0.05).7 Effect of the total grain of defecation in rabbits within 10 h: group A and C,the difference is significant before and after intragastric administration (p<0.01),group B ,the difference is not significant before and after intragastric administrationin (P>0.05).Compare group A with group B and C ,the difference is significant(p<0.01),compare group B with group C, the difference was not significant(P>0.05).8 Effect of the total wet weight of defecation in rabbits within 10 h: group A and B ,the difference is significant before and after intragastric administrationin (p<0.01),group C, the difference is significant before and after intragastric administrationin (p<0.05).Compare one group with another: the difference is significant between group A and group B (p<0.01), the difference is significant between group A and group C(p<0.01), the difference is significant between group B and group C(p<0.01).9 Effect of moisture content of the large intestine of rabbits: There is no significant difference among each group on jejunum water content, ileum water content, colon water content(P>0.05).But on rectum water content, compare one group with another: the difference is significant between group A and group B(p<0.01), the difference is significant between group A and group C(p<0.01), the difference is significant between group B and group C(p<0.01).10 Effect of Stool water content of Stool in rabbits: After intragastric administrationin , group A compare with group B no significant difference (P > 0.05), with group C is significantly different (p <0.01),the difference is significant between group B and group C(p<0.01).11 Effect of blood MTL in rabbits: group A, the difference is significant before and after intragastric administrationin (p<0.01), group B and C, The difference is not significant before and after intragastriafter intragastric administrationin (P>0.05). Compare group A with group B and C, the difference is significant(p<0.01),compare group B with group C, the difference is not significant(P>0.05).12 The effect of serum electrolytes in rabbits: The difference is not significant before and after intragastric administrationin each group (P>0.05). The difference is not significant among the three groups (P>0.05).13 Effect of Hemorheology of rabbits: In whole blood viscosity, Plasma viscosity and Hematocr, the difference is significant before and after intragastric administration in group A(p<0.01), the difference is not significant before and after intragastric administration in group B and C(P>0.05). Compare group A with group B and C, the difference is significant (p<0.01), compare group B with group C, the difference is not significant (P>0.05).Conclusions:1 Zengye chengqi decoction can change the stool form of rabbits, make the defecation smooth and unobstructed, but not lead to diarrhea.2 Zengye chengqi decoction can reduce the time of defecation in rabbits obviously, increase grain and wet weight of defecation, increase the moisture content of stool and rectum.3 Zengye chengqi decoction can improve hemorheology, maintain balance of the blood electrolyte in rabbits.4 Zengye chengqi decoction can increase peristalsis and increase blood MTL level in rabbits,.5 Zengye chengqi decoction is consistent with Forlax on defecation function and no side effects. While Zengye chengqi decoction may improve blomicrocirculation at the same time, it strongly proves that TCM had advantage in holism in clinical treatment, and provides reliable experimental data for treating a variety of constipation. Clinical treatment of constipation can be used as the preferred prescription.
