Effects Of Self-made Prescription Ointment On The Contents Of IL-1? And IL-6 In Acute Closed Soft Tissue Injury In Rats

Objective: To establish a blunt contusion model of acute closed soft tissue in SD rats by means of percussion method,to use self-made prescription as experimental drug,and to select Erhuang analgesic ointment as positive control drug with similar prescription composition,to observe the changes of limb recovery in rats,and to explore its therapeutic effect and mechanism on acute soft tissue injury by detecting the changes of interleukin-1?(IL-1?)and interleukin-6(IL-6)content in injured tissues,To compare the efficacy of Erhuang Xinchang Zhitong Ointment in the treatment of acute soft tissue injury and to provide further research and clinical application of the drug Theoretical basis.Methods :80 male SD rats were taken,A model of acute closed soft tissue injury by strike,The SD rats were randomly divided into four groups(n=18),i.e.A group(self-made normal concentration),B group(self-simplified high concentration),C group(Erhuang Xinchang Zhitong ointment),Group D(Vaseline).Different groups of rats were randomly divided into different phase points group(n=6),After the mold,2h began to apply,Once a day,8 h.each The symptoms of soft tissue injury were observed at different time points in 1,4 and 7 days;Observation and evaluation of histomorphology by light microscope The content of IL-1??IL-6 in tissue homogenate was detected by enzyme reaction(ELISA),and the results were analyzed by SPSS20.0.Results :(1)The scores of injury symptom index were similar on the 1st day.There was no significant difference between the four groups(P>0.05).On the 4th day,the average scores of normal concentration group,high concentration group and analgesic ointment group were the same.There was no significant difference(P>0.05)between the three groups and vaseline group.The difference was PP>.On the 7th day,the injury index of the self-made prescription group,the self-made prescription high concentration group and the Erhuang Xin injury analgesic ointment group was still lower than that of the vaseline group,among which the self-made prescription high concentration group had the lowest value compared with the vaseline group The difference was significant(P<0.05),compared with the normal concentration group and the second yellow new injury analgesic ointment group(P<0.05).(2)Histomorphological observation: Compared with vaseline group,the self-made normal concentration group,the self-made high concentration group and the Erhuang new injury analgesic ointment group could significantly reduce the inflammatory response of local soft tissue after acute soft tissue injury in rats and accelerate the repair of muscle fibers.(3)IL-1? content of tissue homogenate: the IL-1? in the four groups of rats increased first and then decreased at the three time-phase points Potential.The IL-1? values of low concentration group,high concentration group and Erhuang Xinchang Zhitong ointment group were lower than those of Vaseline group at 1,4 and 7.The lowest values of P< high concentration group were significantly different from those of normal concentration group and Erhuang Xinchang Zhitong ointment group(P<0.05).(4)Tissue homogenate interleukin-6(IL-6)content: the IL-6content in the tissue of the four groups reached the peak value on the 3rd,the self-made normal concentration group and the self-made high concentration group There were significant differences between the two groups(P<0.05),the high concentration group and the normal concentration group and the P< group(P<0.05).Conclusion: Self-made prescription ointment can significantly improve the symptoms and signs of acute soft tissue injury rats and promote the recovery of limb function.The normal concentration of self-made prescription is similar to that of Erhuangxin injury analgesic ointment.The effect of self-made prescription on acute soft tissue injury rats is better than that of self-made prescription and Erhuangxin injury analgesic ointment.
