The concentration and composition of plant secondary compounds are well known to confer plant resistance to herbivores,pathogens and mechanical wounding.Glucosinolates(GSs)are a major group of nitrogen-and sulfur-containing secondary metabolites of Arabidopsis thaliana.The rosette leaves of A.thaliana were wounded by cutting approximately 40%leaf area across the apical lamina with scissors,including entire mechanical wounding,half mechanical wounding and part mechanical wounding.The leaves were harvested after the treatment.GSs were extracted from rosette leaves.The dominant GSs were analyzed by HPLC.Protein was extracted from rosette leaves. Differentially-expressed proteins were studied.(1)12 GSs detected in rosette leaves.8 aliphatic GSs,3-methylsulphinylpropyl GS,4-hydroxybutyl GS,4-methylsulphinylbutyl GS,5-methyl-sulphinylpentyl GS,6-methylsulphinylhexyl GS,8-methylsulphinyloctyl GS,7-methylthioheptyl GS,8-methylthiooctyl GS,4 indole GSs,4-hydroxyindol-3-ylmethyl GS,indol-3-ylmethyl GS,4-methoxyindol-3-ylmethyl GS,l-methoxyindol-3-ylmethyl GS.(2)Wounding by scissors tend to increase GS contents.At 3 h after treatment,total GS content, aliphatic GSs and indole GSs content were reached highest.The content of 4-hydroxybutyl GS (40HB),4-methylsulphinylbutyl GS(4MSOB),3-methylsulphinylpropyl GS(3MSOP),indol-3-ylmethyl GS(I3M)and 4-methoxyindol-3-ylmethyl GS(4MTT3M)were also increased significantly at 3 h after wounding.(3)The content of GSs was induced by different ways,after entire mechanical wounding and half mechanical wounding,the content of GSs was increased,and after entire wounding,the content of GSs was increased more than after half wounding.After part mechanical wounding,the content of GSs was decreased.(4)After wounding treated,intensity changes of 58 protein spots were detected with statistical significance.34 protein spots were found responding to 1.5 h,20 protein spots of them up-regulated otherwise 14 down-regulated differentially-expressed proteins were identitied.36 protein spots were found responding to 3 h,23 protein spots of them up-regulated and 13 down-regulated.
