Objective: To replicate the rat model of constipation with yin deficiency and intestinal dryness by using the " Sweating compound diarrhea method ",and to evaluate the macroscopic characterization,microscopic biological indications,and the " Syndrome Differentiation through Formula Effect Assessment" of the model,so as to provide an experimental basis for the study of the rat model of constipation yin deficiency with intestinal dryness.Methods: Eighty rats were randomly divided into blank group,model group,model control group,Qi-deficiency prescription group,blood-deficiency prescription group,yin-deficiency prescription group,yang-deficiency prescription group,heat secret prescription group,10 rats in each group.Rats in the model group and five syndrome groups were induced by compound method of Pilocarpine and Rhein,and rats in the model control group were induced by loperamide for 14 days.At the end of the modeling,each group was treated with corresponding classical prescriptions of Chinese medicine: Huang Qi Tang for Qi-deficiency prescription group,Run Chang Wan for blood-deficiency prescription group,Zeng Ye Tang for yin-deficiency prescription group,Ji Chuan Jian for yang-deficiency prescription group,and Ma Zi Ren Wan for heat secret prescription group,and the treatment lasted for 4 days.The rats were observed for general condition,body weight,food intake,fecal properties,the number of 24 h fecal pellets,5h fecal water content,and the number of fecal pellets in the colon after execution;the mucus secretion of the colon were detected,the serum VIP content was detected by ELISA,the serum NO content was detected by colorimetric method,and the colonic AQP3 and AQP8 expressions were detected by immunohistochemistry.Results:(1)Evaluation of disease model: The coats of the rats in the model group are sparse and moist,the ears,nose,lips,and tail are dull,and the stools are sparse in the early stage of modeling and dry and hard in the later stage.The water content of feces at 5h and the number of fecal particles at 24 h are significantly lower than those of the blank group.And the model control group(P<0.05),the number of fecal particles in the colon was significantly higher than that of the blank group and the model control group(P<0.01).In terms of microscopic indications,the number of goblet cells in the colon tissue and the expression area of positive cells in the model group were significantly lower than those in the blank group and the model control group(P<0.01),serum neurotransmitter(NO,VIP)content,colon The expression of aquaporin(AQP3,AQP8)was significantly higher than that of the blank group(P<0.05);(2)The effects of Zengye decoction on the water content of feces in 5 hours,the number of fecal particles in 24 hours,the number of fecal particles in intestine,the number of goblet cells in colon tissue and the expression area of positive cells,the content of serum neurotransmitters(NO,VIP)and the expression of aquaporins(AQP3,AQP8)in the model rats were better than those of other prescription and syndrome groups.Conclusion: The injection of pilocarpine combined with rhein for 14 days can establish a rat model of constipation with yin deficiency and intestinal dryness.
