Objective:The aim of this study is to investigate the prescription patterns of functional constipation(FC)in Chinese medicine,using data mining technology combined with the research ideas of network pharmacology to analyze its core prescriptions.By analyzing the therapeutic mechanism of FC in Chinese medicine in a comprehensive manner,we aim to provide some reference for the clinical treatment and experiment of FC.Methods:1.The study covered the literature of the past 10 years,which was collected and sorted out by computer searches of databases such as CNKI,Wanfang and Wipu to screen out studies that met the inclusion and exclusion criteria.The Chinese medicine names and group prescriptions were normalized,and then by Excel 2019,IBM SPSS Modeler 18.0 and IBM SPSS Statistics 26.0 software,the data mining methods were used to collate the frequency of medication,the four qi,the five tastes and the attribution respectively,and association rule analysis and cluster analysis techniques were applied in order to screen out the core medication combinations The core combinations of medicines are then analysed to determine the patterns of use in the treatment of FC.2.Based on the TCMSP platform,we searched the components and targets of the drugs,normalized them through the uniport database,used the GeneCard database and OMIM database to retrieve information on the disease targets related to FC,and constructed a disease-Chinese medicine active ingredienttarget network map in cytoscape software.The network diagrams were constructed in cytoscape software,and the intersection of active drug targets and diseases was plotted in Venny.The target information was uploaded to the STRING platform for PPI network mapping,and the core targets were selected.Finally,GO functional enrichment analysis and KEGG pathway enrichment analysis were performed to annotate the role of the active target proteins in gene function and to elucidate the major signalling pathways of the drug for disease treatment.Results:1.After data mining and screening,241 prescriptions and 162 herbal medicines were counted,with a total of 2540 occurrences,of which Atractylodes macrocephala,Glycyrrhiza glabra,Radix Angelicae Sinensis,Fritillariae Sinensis,Citrus astragali,Herb a Ci stanches,Bitter almonds,Citrus aurantium shells,and Radix et Rhizoma rawhodis are the top 10 herbal medicines in terms of frequency of use.The main herbal properties are "warm",the five tastes are "sweet",and the meridians are "spleen meridians".The association rule analysis was used to explore the correlation of drug combinations.45 groups of drug combinations with high support were selected based on the support analysis and grouped into five clusters using cluster analysis.2.Based on the analysis of support and confidence values,the drug combination "Atractylodes macrocephala-Astragali-Angelica sinensis" was characterized by high support and confidence,and network pharmacology analysis was conducted on this group of core drug combinations..The topological analysis was performed and the main compounds in the drug combinations were obtained as quercetin,β-sitosterol and dousterol.After PPI network construction analysis,the core targets of the core drug combinations for FC were mainly distributed in JUN,MAPK1,HSP90AA1,AKT1,etc.GO functional analysis of the biological processes(BP)for FC were mainly enriched in the response to lipopolysaccharide,response to molecules of bacterial origin,etc.;cellular components(CC)were mainly enriched in membrane rafts,membrane micro-regions,etc.;molecular functions(MF KEGG enrichment analysis revealed that the pathways associated with FC treatment are:IL-17 signalling pathway,PI3K-Akt signalling pathway,MAPK signalling pathway,etc.Conclusion:1.Through data mining analysis,it can be seen that the onset of functional constipation is mostly related to the spleen,stomach,lung and large intestine,and in the treatment of functional constipation,most of the medicines are mainly used to nourish deficiency and regulate qi.Therefore,in the treatment,attention should be paid to invigorating the spleen and stomach,and supplemented by the method of promoting Qi and moistening.2.In the network pharmacological analysis," Atractylodes MacrocephalaeAstragalus-Angelicae”is used to treat this disease mainly through the targets of JUN,MAPK1,HSP90AA1,AKT1,RELA,TNF,IL6,FOS,MYC,MAPK14,etc.to regulate IL-17,PI3K-Akt,MAPK,calcium ion,Ras,choline,etc,Calcium ions,Ras,cholinergic synapses,cAMP,dopaminergic synapses,mTOR and other signaling pathways,the mechanism of action is mostly to inhibit inflammation,repair damaged intestines,increase intestinal water,lubricate the intestine,enhance intestinal dynamics and other aspects of regulation,in which the horned medicine“Atractylodes MacrocephalaeAstragalus-Angelicae”plays an important role in the treatment of FC,achieving the function of replenishing the Qi of the spleen and stomach and helping to moisten the stomach.
