| Antibiotics can treat diseases caused by bacteria,mycoplasma,spirochetes and other microorganisms infecting people or animals;It can also be widely used as antibiotic growth promoter in animal husbandry.Feed antibiotics are widely used because they can prevent and cure animal diseases,promote animal growth and improve feed remuneration.However,with the increasingly serious adverse effects of abuse of feed antibiotics on animal health,animal derived food pollution,ecological environment pollution,harm to human health and drug-resistant bacteria,the main animal husbandry countries in the world have adopted the prohibition of feed antibiotics.Pure natural Chinese herbal medicine has become one of the hot research topics to replace feed antibiotics.Florfenicol is one of the most commonly used chloramphenicol antibiotics in broiler farms,and its popularity rate is 78%.The experimental study was conducted to observe the intestinal barrier damage of broiler antibiotic related diarrhea induced by flufenicol,and to observe the prevention and treatment of the classic compound Chinese medicine Shenling Baizhu powder and self-made Chinese medicine,which provides the basis for the research of compound Chinese medicine for anti-inflammatory.methods125 8-day-old healthy Ross broilers were randomly divided into blank group,florfenicol group(drinking water added with 100 mg/kg?bw?d florfenicol aqueous solution)and high-dose compound Chinese medicine group(drinking water added with 100 mg/kg?bw?d florfenicol+2 g/kg?bw?d self-made compound Chinese medicine water decoction),low-dose compound Chinese medicine group(drinking water added with 100mg/kg?bw?d florfenicol+1 g/kg?bw?d self-made compound Chinese medicine water decoction),Shenling Baizhu powder group(drinking water added with 100 mg/kg?bw?d florfenicol+2 g/kg?bw?d Shenling Baizhu powder water).The number of diarrhea of broilers was recorded every day,and the diarrhea rate of broilers during the experiment was counted.The broilers were randomly sampled at 14,21 and 28 days to observe the clinical symptoms and weigh.The pathological changes of jejunum were observed by HE staining method,and the expression of HIF-1α,NOX4,Ang II and tight junction proteins occludin,claudin-1 and ZO-1 were detected by ELISA in 28 days broilers.results1.Compared with the blank group,the broilers in florfenicol group were slow in action,depressed in spirit,drooping in wings,disheveled in feathers,increased in drinking water but decreased in food intake,gradually emaciated,and discharged brown and thin stool;the liver,spleen and intestinal tract were swollen,the liver was yellowish,the intestinal wall became thin and transparent,and there were bleeding points in the intestinal tract.Compared with florfenicol group,the above clinical symptoms and autopsy changes of broilers in high-dose group,low-dose group and Shenling Baizhu powder group were significantly improved.2.Compared with the blank group,the diarrhea rate of broilers in florfenicol group at 8-14 days,15-21 days and 22-28 days was significantly increased(P<0.05);compared with florfenicol group,the diarrhea rate of broilers in high-dose compound Chinese medicine group,low-dose compound Chinese medicine group and Shenling Baizhu powder group was significantly decreased(P<0.01).3.Compared with the blank group,the body weight of broilers in the florfenicol group at 14,21 and 28 days of age was significantly lower(P<0.05),and with the increase of age,the weight loss proportion of broilers in the model group gradually increased,with a minimum of 40%;compared with florfenicol group,the body weight of broilers in high-dose group,low-dose group and Shenling Baizhu powder group at 14,21 and28 days of age were significantly increased(P<0.01),and with the increase of age,the body weight of broilers in high-dose group could be increased by 70%.4.Compared with the blank group,the structure of jejunum mucosa of broilers in florfenicol group was damaged,intestinal villi were arranged disorderly,there were rupture and shedding phenomenon,and the serosa layer of intestinal wall fell off;compared with florfenicol group,the structure of jejunum mucosa of broilers in high-dose group of compound Chinese medicine,low-dose group of compound Chinese medicine and Shenlingbaizhu powder group was basically complete,and the serosa layer of villi and intestinal wall was removed The falling phenomenon was obviously improved.5.Compared with the blank group,the protein contents of HIF-1α,NOX4 and Ang II in the 28 days florfenicol group were significantly increased(P<0.01);compared with the florfenicol group,the protein contents of HIF-1α,NOX4 and Ang II in the jejunum of broilers in the high-dose group of compound Chinese medicine,the low-dose group of compound Chinese medicine and the positive Shenlingbaizhu powder group were significantly decreased(P<0.01).6.Compared with the blank group,the protein contents of ZO-1,occludin and Claudin-1 in the 28 days florfenicol group were significantly decreased(P<0.05);compared with the florfenicol group,the protein contents of ZO-1,occludin and Claudin-1 in the jejunum of broilers in the high-dose group of compound Chinese medicine,the low-dose group of compound Chinese medicine and the Shenling Baizhu powder group were significantly increased(P<0.05).conclusions1.100 mg/kg?bw?d florfenicol solution was continuously fed to broilers for 1-3 weeks,which could lead to diarrhea in broilers.The mechanism of diarrhea in broilers induced by florfenicol was closely related to the damage of jejunal barrier function,as well as the increase of jejunal hypoxia,oxidative stress and inflammation.2.When broilers were fed with 2 g/kg?bw?d,1 g/kg?bw?d and 2 g/kg?bw?d Shenling Baizhu powder for1-3 weeks,it could effectively prevent and treat diarrhea caused by florfenicol in broilers.The mechanism of action was closely related to the effective alleviation of functional damage of jejunal barrier caused by florfenicol,and the improvement of jejunal hypoxia,oxidative stress and inflammatory reaction. |
