| Diarrhea induced by enterotoxigenic Escherichia coli(ETEC) is a communicable disease of newborn piglets and one of the major diseases of serious harm to the swine industry, which often has brought huge economic losses to the pig industry. Therefore, in the modern pig industry, prevention and treatment of diarrhea is solved and must be solved. Currently, due to the long-term use of antibiotics will decrease the sensitivity of bacteria to drugs, have a great resistance and drug residues. In addition, since there are many different serotypes and pili of of E. coli gene, so the application of vaccines difficult to achieve a comprehensive prevention. As a consequence, the development of a new drug prevention and treatment of the disease is imminent.In China, the application of traditional Chinese medicine has had thousands of years of history of the disease.The medicine contains many active ingredients that is antibacterial and can enhance the role of the immune system, and there is no drug residues and other issues. Currently, in clinical, the herb has been used to treat E. coli diarrhea in piglets. Therefore, the application of traditional Chinese medicine in treatment of piglet escherichia coli diarrhea is a good and effective choice. This study aimed to screen out effective prescription drugs to treat the piglet diarrhea and provide a theoretical basis for the future treatment of the disease. Contents and results are as follows:At first,we selected the 28-day-old piglets and enterotoxigenic Escherichia coli diarrhea established a model to determine the optimal amount of poison attack of diarrhea caused by E. coli is 6 × 109 CFU / mL. Find in foreign and domestic literature and then select the 23 kinds of traditional Chinese herbs, and minimum inhibitory concentration(MIC) of E.coli was measured, according to the MIC values of herbs, as well as the principle of incompatibility and the composition of the two pairs of prescription medicine. And then two pairs of prescription and enrofloxacin for the control group performed in vitro experiments showed that the inhibition zone of prescription medicine one is 1.436 ± 0.024 cm, the antibacterial ring of prescriptiontwo is 1.367 ± 0.016 cm,the antibacterial ring of enrofloxacin is 1.313 ± 0.019 cm.On this basis, we fed the traditional Chinese medicine and western medicine for diarrhea piglets according to the groups and tested the treatmet effect of medicines. Experimental designed as follows: twenty 28-day-old piglets were randomly divided into four groups: the blank group(do not feed any drug), the first group of Chinese Herbs, the second Chinese Herbs, enrofloxacin group. Each group was fed piglets serving 5mL of 6 × 109 CFU / mL E. coli bacteria before feeding medicines. After all piglets were diarrhic, we according to the packet feed medication for 10 days,during feeding medication, every other day recorded in each group weight, feed intake, fecal scoring, the cure rate, efficiency, mortality, the number of rows of bacteria. After two hours for feeding medication on the10 th days early in the morning, we collected blood from the precaval vein and detected proliferation and transformation capabilities in peripheral blood lymphocyte and bacteriostatic ring of serum antibacterial experiment. After abataging,we took each bowel of small intestine to HE staining and made pathological sections,then observed histopathological changes in various bowel. The results showed: the average weight and average gain of the first Chinese Herbs group is relatively heavy with the blank group piglets significantly different(P <0.01), the average weight of piglets of the first Chinese medicine prescription is higher than the second medicine prescription and enrofloxacin group.Average daily feed of the first Chinese medicine prescription,the second traditional Chinese medicine prescription group and enrofloxacin group is higher than the blank group. The average daily feed intake of the first Chinese Herbs groug,the second Chinese Herbs group and enrofloxacin group was higher than the blank group, in which the first Chinese Herbs group piglets ADFI compared with the blank group were significantly different(P <0.05).ADFI of the first Chinese Herbs group was higher than the second group and enrofloxacin group.The fecal scoring of the first Chinese Herbs groug,the second Chinese Herbs group and enrofloxacin group is lower than the blank group, in which the first Chinese Herbs piglet fecal scoring compared to the blank group were significantly different(P <0.05). The fecal scoring of the first Chinese Herbs was lower than the second Chinese Herbs group and enrofloxacin group.The cure rate and the effective rate of the first Chinese Herbs group,the second Chinese Herbs group and the enrofloxacin group was higher than the control group and the difference was significant(P<0.05).The cure rate and the effective rate of the first Chinese Herbsgroup was higher than the second Chinese Herbs group and enrofloxacin group. The number of rows of bacteria of the first Chinese Herbs group,the second Chinese Herbs group and the enrofloxacin group was lower than the blank group and the difference was very significan(P <0.01), in which compared the first Chinese Herbs group with the second Chinese Herbs group,the number of rows of bacteria was significant(P <0.05) and the number of the rows of bacteria of the first Chinese Herbs group was lower than the second Chinese Herbs group. In peripheral blood lymphocyte proliferation(SI) of the first Chinese Herbs group was higher than the blank group,the second Chinense Herbs and the enrofloxacin group,in which SI of the Chinese Herbs group compared to the blank group,the difference was extremely significant different(P <0.01). The SI of the second Chinese Herbs group and the enrofloxacin group compared to the control group,the difference was significant(P <0.05). The serum inhibition experiment showed that the inhibition zone of blank group was 0.600 ± 0.000 cm,the inhibition zone of the first Chinese Herbs group was 1.832 ± 0.171 cm,the inhibition zone of the second Chinese Herbs group was 1.625 ± 0.226 cm, the inhibition of the enrofloxacin group was 1.607 ± 0.135 cm.Compared the first Chinese Herbs group to the control group,the difference was extremely significant(P <0.01).Pathologies showed that villus of duodenum and jejunum of the blank group were shedding,submucosa bleeding and increased inflammatory cells.The villus of duodenum and jejunum of the first Chinese Herbs group were integral, midgut gland structure was clear and the nucleus was intact.The midgut gland structure of the duodenum and the jejunum of the second Chinese Herbs group were not clear and increased inflammatory cells.In the enrofloxacin group,part of the midgut gland of the duodenum,part of the intestinal villi had inflammatory cell infiltration and some bleeding phenomenon. Synthesize all datas,the results showed that the first Chinese Herbs and the second Chinese Herbs both can treat the diarrhea induced by E.coli, the first Chinese Herbs is better. |
