Object:To study the protective effect of single Chinese herbal medicine water extract on the intestinal mucosal barrier function of broiler chickens,and to provide basic theoretical data for the prevention and treatment of poultry diseases by traditional Chinese medicine.Method:280 1-day-old male jute chickens were divided into fourteen groups,two control groups,two model groups,ten Chinese medicine groups,20 in each group,and the control group was intraperitoneally injected with physiological saline at the age of 15-17 days.The model group and The Chinese medicine group was intraperitoneally injected with Escherichia coli O₇₈8 at the age of15-17 days.The Chinese medicine group started to drink Chinese medicine at the 18th day of age.For 7 consecutive days,each chicken has 10ml of Chinese medicine liquid with a concentration of0.02g/mL per day?the Chinese medicine liquid contains 1g of Chinese medicine per ml?,and it is consumed within two hours.Blood was collected on the 7th and 14th days after the administration,and the contents of serum diamine oxidase,D-lactic acid and endotoxin were measured by ELISA method,the small intestine segment was collected,the intestinal coefficient was calculated,and tissue sections were prepared to observe the intestinal morphology and structure.The real-time fluorescence quantitative PCR was used to detect the mRNA expression of tight junction proteins ZO-1,Claudin-1 and Occludin to explore the protective effect of Chinese medicine water extract on the mechanical barrier of the small intestine;The DNA of cecum contents was sequenced by16S rDNA high-throughput detection technology to study the effect of single Chinese medicine water extract on the structure and quantity of cecum flora in the intestinal biological barrier;By calculating the immune organ index,ELSA method was used to detect the serum IgA,IgM,total protein,albumin and intestinal mucosa sIgA content,paraffin section-HE staining method was used to detect the number of intestinal mucosal immune cells to explore the effect of Chinese medicine water extract on the immune function of intestinal mucosa.Result:?1?On the 7th day after
