Objective: Diarrhea is a common digestive disease in canine,and its etiology is complex,including gastrointestinal dysfunction,viral or bacterial infections,etc.The treatment of this prescription is mainly relieve diarrhea with astringents and tonify qi and deficiency.Medicine terminalia fruit nutmeg has the effect of astringent bowel and diarrhea,and can be used for the symptoms of chronic diarrhea and dysentery caused by spleen and kidney deficiency and cold,incontinence of defecation.Qi-enriching drugs are often used in the treatment of diarrhea in canine,Spleen and stomach meridian,has a good effect on spleen deficiency.This article uses the senna leaf-induced diarrhea model to study the anti-diarrheal effect of compound Chinese medicine on senna-induced diarrhea dogs and the mechanism of aquaporin regulation,aiming to explore the therapeutic effect of compound Chinese medicine on diarrhea,provides experimental data for the further application of compound Chinese medicine in small animals.1.Senna leaf feeding was used to establish a diarrhea model for dogs.The test was divided into a control group and a diarrhea model group.The diarrhea model group was fed with 2 g/kg body weight of senna leaves in canine.The effects of clinical signs,defecation times,fecal characteristics,diarrhea rate and diarrhea index on diarrhea caused by senna leaf were evaluated in canine.The results showed that in the senna-induced diarrhea dog test,the number of bowel movements and fecal traits in the diarrhea model group were significantly higher than the control group,and the diarrhea rate and diarrhea index were significantly higher than the control group,the diarrhea model of senna-induced diarrhea dogs was successfully established.2.Observation of the therapeutic effect of Chinese herbal compound preparations on diarrhea dogs.16 healthy dogs were randomly divided into 4 groups after the 7-day adaptability test: A,B,C,D groups,4 in each group.2 g/kg body weight was fed to senna leaves to establish a diarrhea model for dogs,group A was not treated;group C was a traditional Chinese medicine treatment group;Group D is a Western medicine treatment group;group B was no treatment group.The clinical manifestations and symptoms,fecal nature and morphology of the dogs in each group were observed throughout the trial and data were recorded according to the Bristol stool classification method.On 7 days,the anterior colon of the dogs in groups A,B,and C was intercepted by surgery,and the changes of tissue structure and the distribution and expression of the immunohistochemical positive products of AQP3 and AQP4 were studied by HE staining and immunohistochemical staining,respectively.The research results are as follows:(1)Clinical performance results: On the second day after feeding Chinese medicine,the number of diarrhea was significantly reduced,appetite increased,and the spirit improved;on the third day,both the spirit and appetite recovered,and the cure rate was 100%.At the third day after treatment with gentamicin sulfate,the frequency of diarrhea was reduced,and the cure rate was 75%.Fecal trait score: The fecal traits of groups B,C,and D all changed significantly on the third day of the test.Compared with group A,the remaining 3 groups of Bristol stool scores Both were significantly increased(p <0.05);on days 4-6,compared with group A,the Bristol fecal score in group B was significantly increased(p <0.05),and the difference between groups C and D was not statistically significant.(2)Pathological and anatomical changes: The colon structure of group B was loose,the intestinal villi were damaged,the integrity was reduced,the villi interstitium was widened,and the lymph follicles were formed with hollows.The intestinal villi of the tissue are intact,the interstitial villi are tight,the structure is clear,the structure of the lymphatic follicles is tight,and no voids are seen in group C.(3)Changes in protein expression of AQP3 and AQP4 in the colon: Compared with group A,The AQP3 immunopositive products in the colon were significantly reduced in the untreated diarrhea model group B,and after treatment with traditional Chinese medicine,the protein expressio n of AQP3 in the colon was significantly increased.The protein expression of AQP4 was not statistically significant.The test proves that the compound Chinese medicine in this study has an obvious anti-diarrheal effect on dogs with senna-induced diarrhea,and the therapeutic effect is better than that of intramuscular injection of gentamicin sulfate injection.The compound Chinese medicine developed in this study plays an anti-diarrhea effect by inhibiting the decrease of AQP3 expression.
