With the increase of people's demand for computers,complex problems appear which are difficult to be solved by traditional computers.First,with the increase of the scale of information processing,the demand for computer data storage increases.Second,the recent shortage of chips around the world also makes us realize the importance of improving the speed of chip making.DNA computing is a new technology to solve this problem.The permutations and combinations of four DNA bases expand the information storage capacity,and the self-assembly and displacement functions of DNA strands could complete the production of DNA chips quickly.DNA logic chip is an important part of DNA computing.DNA chips contain chip holders,DNA probes,and the latter is cured and connected to the surface of the former.Depending on the holders,DNA chips are divided into DNA inorganic chips and DNA organic chips.The commonly used holders for DNA inorganic chips are silicon,glass,and so on;the commonly used holders for DNA organic chips are nylon membrane,polypropylene screen,etc.In this paper,with the help of caDNAno,NUPACK and other designs,the design of chip holders with DNA as the material is finished through the DNA assembly,which belongs to the innovation of DNA organic chip holders;the DNA logical circuit is successfully designed by encoding the DNA logical computing model into DNA strands.Finally,the DNA logic chip has been realized by curing and connecting the DNA logical circuit to the DNA holders.Agar gelatin electrophoresis,Native-PAGE and Real-time PCR have been used to investigate various aspects of the logic chip.The first is the holder of DNA chip,We studied the existing organic holder,including membrane vesicles,dcDNA-Ti3C2,and so on,and proposed DNA origami was used as holder DNA logic chip.Based on DNA assembly technology,the design of DNA chip holders have been realized by using M13mp18 and DNA staples.Through this work,we have explored DNA holders of DNA chip,making the DNA chip materials unified and with a higher integration.The second is using DNA rings as the basic functional strands,which is unfolded in the presence of input strands,and the unfolded DNA rings exposes the hidden region,displacing the reporter strands.Based on this,the design of the "yes","and" and "no" logic gates and the DNA cascode circuit are realized.Finally,the functional strands involved in the DNA logical circuit are cured and connected by designing the positioning strands at a more central position of the holders.It's easier than traditional connection by Specially made holes.DNA has a natural of self-assembly,and a variety of nanostructures are formed as a result,which have the characteristics of designability,operability and programming and provide a reference template for DNA chip.DNA logic chip is both the technical basis for molecular computing and also has practical significance for organisms with wide prospect of application.
