| Objective:Recurrent spontaneous abortion(RSA)is a common and difficult disease of Obstetrics and Gynecology.For patients and their families to bring immeasurable psychological blow and economic burden.Serious impact on family harmony.ZhangZhongjing’s "Neijing" first put forward the theory of "kidney deficiency and blood stasis".It provides a good theoretical basis for the treatment of unexplained recurrent spontaneous abortion(URSA),is widely used in clinical treatment of traditional Chinese Medicine.In this clinical experiment,we studied the clinical effect of Bushen Huoxue prescription(URSA)on interleukin-2(Interleukin-2,IL-2)and interleukin-6(Interleukin-6,IL-6).To observe the expression of IL-2Rα、IL-2Rβ、IL-6Rα in decidua of normal early pregnant women and patients with URSA and the content of peripheral blood.The effect of Bushen Huoxue Prescription on 60 cases of kidney deficiency and blood stasis type URSA.To:study the regulation mechanism of Bushen Huoxue prescription on IL-2 and IL-6.More important is to explore the relevance of URSA and IL-2,IL-6.The purpose of this study was to investigate the etiology,pathogenesis and treatment of URSA.Objective to investigate the clinical effect of Bushen Huoxue prescription in preventing and treating URSA,Can improve the pregnancy outcome of URSA patients and improve the success rate of pregnancy,to provide scientific basis for the prevention and treatment of URSA.Methods:This study is divided into two parts,the first part of the collection of normal outpatient artificial abortion decidua in 15 cases as control group,and URSA in patients with failed pregnancy decidua abortion 15 cases as experimental group,using real-time quantitative PCR(RT-PCR)method for the determination of two groups in Decidua Tissue of IL-2Rα,IL-2Rβ,IL-6Rα mRNA content.60 patients with unexplained recurrent spontaneous abortion second cases collected outpatient(6-8 gestational weeks)as the research object,divided into Bushen Huoxue Decoction Combined with dydrogesterone and 30 cases in the experimental group and single dydrogestrone,30 cases in the control group for 4 weeks.After 4 weeks of admission and admission,two groups of patients were collected venous blood,ELISA method was used to detect the serum levels of IL-2 and IL-6 in two groups,and the changes of serum progesterone,estrogen andβ-HCG levels were observed before and after treatment in the two groups.Results:(1)The expression of IL-2Rα and IL-2Rβ in decidual tissues of patients with URSA was significantly higher than that in normal group,and there was significant statistical difference(P<0.05),The expression of IL-6Rα in decidual tissues of patients with URSA was significantly lower than that in normal group,the two groups were statistically significant(P<0.05).(2)The content of IL-2 in the peripheral blood of patients in URSA group and control group was lower than that before treatment,and there was significant statistical difference(P<0.01).The expression of IL-6 in the peripheral blood of patients in URSA group and control group was higher than that before treatment,and there was significant statistical difference(P<0.01).(3)The level of IL-2 in the URSA group was lower than that of the control group after URSA,and there was significant statistical difference(P<0.01).The level of IL-6 in the URSA group was higher than that of the URSA group,the two groups were statistically significant(P<0.01).(4)In the experimental group and the control group,the levels of progesterone,estradiol and chorionic gonadotropin in peripheral blood after 4 weeks of treatment were significantly higher than those before treatment,the two groups were statistically significant(P<0.01).The levels of progesterone,estradiol and chorionic gonadotropin in the peripheral blood of the experimental group were significantly higher than those in the control group after 4 weeks,the two groups were statistically significant(P<0.01).Conclusion:(1)High expression of IL-2Rα and IL-2Rβ in decidual tissues of URSA patients,low expression of IL-6Rα,The results indicated that IL-2 and IL-6 cytokines play a role in the decidual tissues of patients with URSA and normal early pregnancy,It is suggested that IL-2 and IL-6 may be involved in the pathogenesis of URSA.(2)Bushen huoxue prescription may reduce the expression of IL-2 in peripheral blood of patients,increased expression of IL-6,to improve the clinical efficacy of unexplained recurrent spontaneous abortion.(3)Bushen huoxue prescription can significantly improve the URSA patients peripheral blood P,E2,HCG content,to provide the basis for continued normal pregnancy. |
