Clinical Research On Effect Of Bushen Huoxue Recipe On IL23、IL27 Of Peripheral Blood In URSA

Objective：1.To compare the expression of Interleukin-23(IL-23)receptor and Interleukin-27(IL-27)receptor between normal pregnant women’s and unexplain recurrent spontaneous abortion(URSA)patients’ decidua and chorionic tissus.2.To compare the IL-23 and IL-27 levels change in peripheral blood before and after Bushen Huoxue prescription,to explore the correlation between IL-23、IL-27 and URSA.3.To compare the difference of IL-23 and IL-27 in URSA patients’ peripheral blood between after taken Bushen Huoxue prescription plus dydrogesterone and only after taken dydrogesterone.4.To discuss the association of cytokine IL-23 and IL-27 levels in patients with URSA.To study the clinical significance of serum cytokine IL-23 and IL-27 levels in patients with URSA after taken Bushen Huoxue prescription.To provide further theoretical basis for further study of Bushen Huoxue Prescription in clinical treatment of URSA patients.Methods:In this study,subjects were divided into two parts.1.Part 1 collects 15 cases of abortion decidua in artificial abortion patients(6-10weeks of pregnancy)and 15 cases of abortion decidua in patients with URSA(6-10weeks of pregnancy)with the methods of RT-PCR to determine the levels of IL-23 receptor and IL-27 receptor.2.Part 2 collects 60 cases of patients with URSA(6-8weeks of pregnancy).60 patients were divided into two groups by random,URSA group consisted 30 cases and artificial abortion patients group consisted 30 cases.The patients were randomized either to give dydrogesterone or to give Bushen Huoxue prescription plus dydrogesterone for 4 weeks of treatment.Collect their blood 4-5ml when they are in and out of our study.Use ELISA to determine the change of IL-23 and IL-27 levels,and to monitor the patients of two groups serum progesterone(P),estradiol(E2)and β-HCG levels before and after treatment.Result:1.The patients with URSA IL-27 receptor mRNA content in deciduas are lower than Normal abortion patients,and IL-23 receptor content in deciduas are higher than Normal abortion patients.2.The content of IL-23 in the peripheral blood of patients with URSA in the experimental group treated by the traditional Chinese medicine Bushen Huoxue Recipe and Western medicine was significantly lower than that before treatment,and the difference was statistically significant(P<0.01).The content of IL-23 in peripheral blood of URSA patients treated by Bushen Huoxue Decoction and Western medicine dydrogesterone in the experimental group was significantly lower than that of the control group were treated with western medicine dydrogesterone,and the comparison between the two groups was statistically significant(P<0.01).3.The content of IL-27 in the peripheral blood of patients with URSA in the experimental group treated by the traditional Chinese medicine Bushen Huoxue Recipe and Western medicine was significantly higher than that before treatment,and the difference was statistically significant(P<0.01).The content of IL-27 in peripheral blood of URSA patients treated by Bushen Huoxue Decoction and Western medicine dydrogesterone in the experimental group was significantly higher than that of the control group were treated with western medicine dydrogesterone,and the comparison between the two groups was statistically significant(P<0.01).4.The content of progesterone,estradiol and β-HCG in peripheral blood of URSA patients treated by Bushen Huoxue recipe was significantly higher than that in control group,and the difference between the two groups was statistically significant(P<0.01).Conclusion:1.The expression of IL-23 receptor and the decrease of IL-27 receptor expression in decidual tissue of URSA patients decreased the embryo implantation,development and embryo implantation,which leads to the failure of pregnancy and abortion.2.Bushen Huoxue recipe could improve the clinical efficacy of IL-27 in patients with unexplained recurrent spontaneous abortion by reducing the expression of IL-23 in peripheral blood and increasing the expression of peripheral blood.3.Bushen huoxue prescription can obviously improve the content of progesterone and estradiol in peripheral blood of patients with URSA,so as to maintain pregnancy.
