Objective: To observe the effects of Yinxieping Pill on the expression levels of p21 and p63 in skin lesions of psoriasis-like mice model induced by imiquimod(IMQ)cream.Methods: 48 albino lab(BALB/c)female mice were randomly divided into normal control group,model control group,Acitretin capsule group,and Yinxieping high,middle and low dose groups,with 8 cases in each.The back hair of mice was removed to form 2cm× 3 cm exposed skin areas.Normal control group was coated with white petrolatum ointment once a day and 62.5mg a time;while other5 groups with 5% IMQ to induce psoriasis Disease model for 8consecutive days,once a day,62.5mg a time.At the same time,mice in each group were given topical medicines by gavage on a daily basis: normal control group and model control group with 0.9%physiological saline 0.4ml,Acitretin capsule group with 0.4ml Acitretin solution,and high,medium and low dose groups of Yinxieping pills with 0.4ml Yinxieping pills at corresponding concentration for 8 consecutive days,once a day.24 hours after the last administration,the skin lesions of mice were observed and photographed for scores of skin lesion areas and PASI;light microscopy was used to observe morphological changes,immunohistochemistry,pathological analysis of HE staining,and RT-PCR,so as to detect the expression levels of p21 and p63 in skin lesions of mice.Results:(1)Compared with normal control group,PASI of model control group had a significant increase.Compared with model control group,PASI scores of Acitretin capsule group and Yinxieping Pill low,medium and high dose groups significantly reduced,with statistically significant differences.(P <0.05)(2)The histopathological changes of skin lesions in Yinxieping Pill low,medium and high dose groups were lighter than those of model control group,and their degree further decreased as dose increased;(3)Compared with normal control group,the immunohistochemical expression and relative mRNA expression of p21 and p63 in model control group significantly increased,with statistically significant differences(P<0.05).Compared with model control group,the immunohistochemical expression of p21 and p63 and relative mRNA expression of Acitretin capsule group and Yinxieping pill low,medium and high dose groups significantly reduced,and as the concentration of Yinxieping pill solution increased,its expression level further reduced,with statistically significant differeces.(P <0.05)Conclusion:(1)Yinxie Ping pill can significantly reduce the PASI score of psoriatic-like mice model skin lesions and work well in the treatment and improvement of psoriatic lesions.(2)The levels of P21 and P63 immunohistochemistry and relative mRNA expression in psoriatic-like mice model skin lesions in model group were significantly higher than those of normal group,which prove that the occurrence of psoriasis may be related to P21 and P63.(3)Since yinxieping pills can reduce the content of P21 and P63 in skin lesions of psoriasis-like mice,it may be one of the mechanisms by which Yinxie Ping pill can effectively treat psoriasis.
