Objective: To observe the effect which Yinxieping pills on psoriasis-like BALB/c mice models in skin lesions,histopathology,transforming growth factor beta as well as interleukin-10 expression and to explore the possible mechanism of the Yinxieping pills on treating psoriasis.Methods:Forty-eight healthful male BALB/c mice were randomly divided into6 groups which respectively were blank group,model control group,Tripterygium wilfordii group,Yinxieping Pills high,middle and low dose groups,eight mice in each group.In addition,4 mice of the same batch are using for pre experiment first.Until the model successfully constructed,the experiment begins.After all the mice were depilated on the back,the back skin were given with imiquimod cream except the blank group with vaseline,1 times /d,for 8 days.At the same time,the corresponding drugs were given by gavage.The blank group and the model control group mice were given physiological saline by gavage.Tripterygium wilfordii group,Yinxieping Pills high,middle and low dose groups mice were given the corresponding drug solution by gavage 1 times /d,for 8 days.After the last administration for 24 hours,the psoriasis area and severity index(PASI)score of the skin in the back of mice was observed;The levels of TGF-β and IL-10 in serum were detected by ELISA method.The skin tissue of the back were taken as pathological sections to compare the histopathology.And in the whole experiment,record the general situation of mice.Results:(1)The PASI score of the skin in the back of psoriasis-like BALB/c mice of Yinxieping Pills high,middle,low dose groups,tripterygium wilfordii group and model control group compared with blank group had significant differences(P<0.05).The PASI score of the skin in the back of psoriasis-like BALB/c mice of Yinxieping Pills high,middle,low dose groups and tripterygium wilfordii group compared with model control group had significant differences(P<0.05).(2)The histopathological changes in skin lesions of Yinxieping Pills high,middle and low dose groups to varying degrees to lighter than the model control group.(3)The content of TGF-beta and IL-10 in Yinxieping Pills high,middle,low dose groups,tripterygium wilfordii group and model control group mice’s peripheral blood was significantly higher than the blank group(P < 0.05).The content of TGF-βin Yinxieping Pills high,middle,low dose groups and tripterygium wilfordii group mice’s peripheral blood was significantly lower than the model control group(P <0.05),the difference was statistically significant.The content of IL-10 in Yinxieping Pills high,middle,low dose groups and tripterygium wilfordii group mice’s peripheralblood was significantly lower than the model control group(P < 0.05),the difference was statistically significant.Conclusion:(1)Yinxieping pilsl can reduce the PASI score of psoriasis-like mice models in skin lesions,and it can improve psoriatic skin lesions.(2)The content of TGF-β and IL-10 in model control group mice’s peripheral blood was significantly higher than the blank group,it shows that TGF-beta and IL-10 in peripheral blood are associated with the pathogenesis of psoriasis.(3)Yinxieping Pills can reduce the content of TGF-β and IL-10 in psoriasis-like mice’s peripheral blood,it is probably one of the mechanisms of Yinxieping pills in the treatment of psoriasis. |
