| Objective: To describe the relationship between Yinxieping Pill and the expression of Notch1 and Caspase3 in mouse skin lesions,and to explore the possible mechanism of Yinxieping Pill in the treatment of psoriasis.Methods:48BALB/c female mice were divided into 6 groups by random method,with 8 mice in each group.The normal group,model control group,acitretin capsule group,Yinxieping Pills low,middle and high dose group.All of the mice were extremely shaved on the back.Medical vaseline was applied externally on the skin of the mice in the normal group,and 5% Imiquimod cream was applied externally on the skin of the mice in the remaining experimental groups.At the same time,the mice in each group were intragastric administration.The normal group and the model control group were treated with normal saline.The acitretin capsule group,Yinxieping Pills low,middle and high dose group were treated with the corresponding drug solution.each group was intragastric administration once a day for 8 days.Observe the general condition of the mice during the experiment.Twenty-four hours after the last intragastric administration of the mice,observed and recorded the Psoriasis Area and Severity Index on the back of the mouse;taken the skin tissue from the back of the mouse and observed the pathological changes under light microscope.The m RNA and proteins of Notch1,Caspase3 were detected by RT-PCR and immunohistochemical SABC method.Results:(1)Compared with the normal group,the model control group,Yinxieping Pills low,middle and high dose group and acitretin capsule group had significant differences in the Psoriasis Area and Severity Index(P<0.05);compared with the model group,the mice in Yinxieping Pills low,middle and high dose group and acitretin capsule group were significantly different in the Psoriasis Area and Severity Index(P <0.05).(2)Compared with the model control group,The histopathologic changes in skin lesions of Yinxieping Pills high dose,middle dose and low dose groups were significantly improved.(3)Compared with the normal group,The m RNA level and protein level of Notch1 and Caspase3 of the model control group were significantly increased there were significant differences between them(P<0.05).The m RNA level and protein level of Notch1 and Caspase3 were much reduced in Yinxieping Pills low,middle and high dose group and acitretin capsule group than model control group,there were significant differences between them(P<0.05).In addition,with the increase of dosage,the expression levels of protein in the low,medium and high dose groups of Yinxieping Pill decreased synchronously,and the difference was statistically significant(P <0.05).conclusion: 1.Yinxieping pills could reduced PASI scores in mice,that Yinxieping pills could improved the symptoms of psoriasis skin lesions.2.The m RNA level and protein level of Notch1 and Caspase3 of the model control group were significantly increased suggested that may be involved in the pathogenesis of psoriasis 3.The m RNA level and protein level of Notch1 and Caspase3 were decreased significantly in the low,medium and high dose groups of Yinxieping Pill,which may be through the regulation of Notch1 and Caspase3 expression to achieve the treatment of psoriasis purpose. |
