Objective:By observing the changes in the expression of IL-32 and IL-35 in the serum of BALB/c psoriasis-like mice after using Yinxieping Pills,to explore the therapeutic effect of Yinxieping Pills on psoriasis.Methods:According to the random number table method,48 sterile BALB/c female mice were equally divided into 6 groups,including blank control group,model control group,Acitretin capsule group,Yinxieping pill low-dose,medium-dose and high-dose groups.Before the experiment,the back of each mouse in each group was depilated to fully expose the skin.The mice in the blank control group were smeared with white petrolatum on their back exposed skin every day,and mice in other groups were smeared with 5% imiquimod cream on their back exposed skin,once a day for 8consecutive days.The wipes were given to each group of mice at the same time by gavage with a gavage needle.The blank control group and the model control group were gavaged with 0.4ml of normal saline by gavage needle,and the acitretin capsule group was gavaged with acitretin A via gavage needle.The drug solution was 0.4ml,and the Yinxieping pill low,medium,and high-dose groups were respectively diluted with the corresponding dose concentration of Yinxieping pill,once a day for 8consecutive days.During the experiment,the back skin lesions of each group of mice were photographed and archived every day.24 hours after the last medication,observe the changes in the skin lesions of each group of mice and give them a PASI score.Take blood from the eyeball to save 1.0ml of blood for each mouse,and use the ELISA method to detect the expression levels of IL-32 and IL-35 in mouse serum.Subsequently,the skin lesions of psoriasis mice were cut and sliced and HE stained to observe the pathological changes.Results:(1)Compared with the blank control group,the severity of the back skin lesions of the mice in the Acitretin capsule group,the low-dose,middle-dose and high-dose groups of Yinxieping Pills,and the model control group were significantly increased by calculating the PASI score(P< 0.05);Compared with the model control group,the severity of back skin lesions in the low-dose,middle-dose,and high-dose groups of the Acitretin A capsule group,the Yinxieping pill group,and the high-dose group were significantly reduced by calculating the PASI score.(P< 0.05);(2)By observing under the electron microscope of HE staining,compared with the model control group,the low-dose,middle-dose and high-dose groups of Yinxieping Pills,and the Acitretin capsule group,the pathological damage of the psoriasis skin tissues were reduced to varying degrees.(3)Compared with the blank control group,the expression of IL-32 in the peripheral blood of mice in the Acitretin capsule group,Yinxieping pill low-dose,middle-dose and high-dose groups,and the model control group all increased significantly(P< 0.05),the expression of IL-35 was significantly reduced(P< 0.05),and the difference was statistically significant;Compared with the model control group,the expression of IL32 in peripheral blood of mice in the Acitretin capsule group,the low-dose,middle-dose and high-dose groups of Yinxieping Pills were significantly reduced(P< 0.05)),and the expression of IL-35 were all Significantly increased(P< 0.05),the difference was statistically significant.Conclusion:(1)Yinxieping pills can improve the PASI score of mice with psoriasis,which suggests that Yinxieping pills have a good effect on the treatment of psoriasis;(2)The expression of IL-32 in the peripheral blood of psoriasis mice in the model control group was significantly higher than that of the blank control group,suggesting that IL-32 may be involved in the pathogenesis of psoriasis;The expression of IL-35 in the peripheral blood of psoriasis mice in the model control group was significantly lower than that in the blank control group,suggesting that IL-35 may play a therapeutic role in the course of psoriasis;(3)The mechanism of Yinxieping Pills for the treatment of psoriasis may be to reduce the expression level of IL-32 and increase the expression level of IL-35 in the peripheral blood of mice with psoriasis.
