The Effect Of The Method Of Invigorating The Kidney And Adjusting The Bubble Cycle On The Ovarian Reproductive Endocrine Function Of PCO Rats

Objective:To observe the effect of Bushen-follicle recuperation therapy;on sexual cycle,endocrine hormones and pregnancy outcome in PCO model rats,and to explore its mechanism.Methods:110 SD rats with regular sexual cycles were randomly divided into eleven groups: blank group,model group,clomiphene positive control group,A-Xiaoyao San(high and low dosages)groups,B-Sanzi Decoction(high and low dosages)groups,Sequential therapy of A+B-Xiaoyao San+Sanzi Decoction in 4 Groups,in each group with 10 rats.PCO model rats were established by drenching letrozole solution.The sexual cycles of PCO model rats were observed before and after treatment.The changes of body masses and pregnant rates were analyzed after treatment.The levels of testosterone,luteinizing hormone,estradiol,progestogen,follicle-stimulating hormone in peripheral serum were measured by enzyme-linked immunosorbent assay.Results: Bushen-follicle recuperation therapy can improve the disordered sexual cycle of PCO model rats,the body mass gains was decreased(P<0.05),The levels of T and LH in peripheral blood of PCO model rats were significantly decreased(P<0.01),the number of embryo of PCO model rats was significantly increased(P<0.01),the pregnancy rate was increased,and the reproductive ability of PCO model rats was improved.The effects of A and B drugs on ovarian function of PCO rats were not significantly correlated with dosages,and no significant interaction was found in the orthogonal experiment.Conclusion: Bushen-follicle recuperation therapy can restore the sexual cycle of PCO model rats,reduce the mass gain,regulate reproductive endocrine,regulate ovarian function,consequently improve pregnancy outcome.However,further experiments are needed to expand the sample size or to demonstrate the interaction and dose-effect relationship of drug combinations by gene and WB detection.
