Objective: the basic theory of traditional Chinese medicine as the basis,combined withmodern medical science and technology, through animal experiment, study of BushenTiaojing Decoction on Poretsky (Neutrul Insulin Injection+human chorionic gonadotrophinneedle subcutaneous injection)induced polycystic ovarian syndrome model rats insulin,fasting (INS)influence the number and ovarian granulosa cells the mechanism andprinciple,discuss Tiaojing Decoction in the treatment of polycystic ovarysyndrome of traditionalChinese medicine of tonifying kidney, providing scientific basis for the effectiveness ofTCM treatment of polycystic ovariansyndrome.Method: This paper selects SPF female rats of70days of age70as the research object,firstly, adaptive feeding for3days, and then under the microscope to observe the changes ofrat vaginal cytology of10days,excluding the10rats were not intact estrous cycle.60ratswere involved in the experiment in the blank control group, normal saline12daily injectionsof the same volume, the other rats by Poretsky method in patients with polycystic ovarysyndrome rat model. In twenty-third days out of8random,detecting vaginal smears, and theblank group2were selected randomly tocontrol, to verify the model making success. In themodel of production is successful, the rats were randomly divided into: model group (modelgroup),Bushen Tiaojing Decoction high dose group (high dose group), BushenTiaojingDecoction low dose group (low dose group), western medicine group,10rats in each group.Under the same feeding conditions, treatment at the same time, the normal group, modelgroup, each rat daily by20ml/kgperfusion of distilled water, high dosage group according tothe daily38.0g//kg Ig; low dose group according to the daily19.0g/kg orally, the westernmedicine group according to the daily0.42g/kg orally metformin, was administeredcontinuously for21days. Weight changes of the rats, the rats were killed, serum level ofFINS free detection method; to observe the changes of ovarian granular cell layer of the lightmicroscope. Results:(1) effect on the weight of rats: after weight data were statistically processed ratsdisplay, model control group was significantly elevatedlevels of body weight, body weight ofrats in each treatment groupdecreased, especially in the high dose group of body weight ofratsdecreased most significantly (P <0.01).(2) on serum insulin (FINS) in rats: effects of statistically treated rats serum FINS contentdata model group (FINS), insulin levels significantly increased,the serum insulin of rats inall treatment groups (FINS) level was decreased,the serum insulin therapy and high dosegroup (FINS) and reduce the level of the most significant (P <0.01).(3) on experimental rat ovarian granulosa cells: normal group rats ovarian granulosa cells for8-9layer, morphological integrity, neatly arranged closely. Ovarian granulosa cell layers ofmodel group rats only1-2layer,and sparse arrangement. Bushen Tiaojing Decoction highdose group of ovarian granulosa cell average4-6layer, forms, arranged more closely.BushenTiaojing Decoction low dose group of ovarian granulosa cell for the3-5layer, form the basicintegrity, arrange disorderly slightly. Metformingroup of the granular cell layer of about3-5layer, form the basic integrity,arrange disorderly slightly.Conclusion: Bushen Tiaojing decoction can regulate the hypothalamicpituitary ovarian axisfunction tends to be stable, reduce weight and ovariancoefficient of rats, decrease theKaohsiung hormones, reduce the highinsulin, improve tissue morphology and structure ofovary, especially theultrastructure, so as to promote the normal development of follicle,ovulation induction success.
