| Objective:To explore the effect of Danzhi Xiaoyao Powder Decoction on the polycystic ovarian rats with the local ovarian granulose cells expression of follicle stimulating hormone receptor(FSHR)and luteinizing hormone receptor(LHR)protein by construct PCO rat model,thus investigating the relevant action mechanisim and its significance of Danzhi Xiaoyao Powder from the aspect of PCOS.Methods:60 female 23-day-old SD rats were randomly divided into two groups.20 SD rats were in normal group.40 SD rats were in model group which were received continuous subcutaneous injection of dehydroepiandrostexone(DHEA).After constucting the model in PCO rats successfully,the selected rats were divided in model group,Danzhi Xiaoyao Powder group and Diane-35 group.Distilled water,Danzhi Xiaoyao Powder and Diane-35 were chronically administered to rats for 21 days,stopped for 7 days for a cycle.3 cycles of administering rats were executed.The serum level of serum sex hormone were measured withradioimmunoassy.Thelocalovariangranulosecells expression of FSHR and LHR protein in PCO rats were measured with immunohistomchemical.Results:(1)PCO rat model assessment:After constructing the PCO model,the body weight in the model group were increased compared with the normal group(P<0.01).Most model rats oestrous cycle were in estrogen interphase which means with anovulation.Model rats ovary presented sack-like structure and follicles-expansive change under light microscope.Compared with the normal group,the serum of T in the model group were increased(P<0.01),moreover the serum of E2 were decreased(P<0.01).(2)The serum of T,FSH and LH changes in PCO rats after drug intervention:Compared with the normal group,the serum of T in the model group,Danzhi Xiaoyao Powder group and Diane-35group were increased significantly(P<0.01).Compared with the model group,the serum of T in the Danzhi Xiaoyao Powder group and Diane-35 group were decreased(P<0.01).Compared with the model group,the serum of FSH in the Danzhi Xiaoyao Powder group were increased slightly,however,the serum of LH were decreased slightly.There was no significant difference in 4 group with the serum of FSH and LH(P>0.05).(3)The local ovarian granulose cells expression of FSHR and LHR protein changes after drug intervention:Compared with the normal group,the local ovarian granulose cells expression of FSHR and LHR protein in the model group,Danzhi Xiaoyao Powder group and Diane-35 group were decreased(P<0.01 or P<0.05).The local ovarian granulose cells expression of FSHR and LHR protein in the Danzhi Xiaoyao Powder group and Diane-35 group were increased compared with the model group after drug intervention(P<0.01).Conclusion:Danzhi Xiaoyao Powder can intensify the expression of FSHR and LHR protein in PCO rats ovarian granulosa cells and lowerthe serum of T which maybe involved inlowering testosteroneand regulating the localovarian granulose cells expression of FSHR and LHR protein. |
