With the explosive growth of information,computers need to process more and more information.However,the technology of silicon chips is difficult to support the increasing amount of computing,which had led to the development bottlenecks in the computer field.To solve this problem,scientists have proposed the concept of biological computer.With the gradual deepening of bio-computer research,people have found that the parallel computing power of DNA,nano-molecular structure,and ultra-high storage capacity have great advantages in breaking through the bottleneck of computer development.Therefore,biomolecular logic operations,such as DNA computing or DNA programming diagnostics,have received widespread attention.At present,most biomolecular logic operation models are detected by fluorescence.Unfortunately,fluorescence detection usually has complicated processing procedures and lacks detection portability.Among number of alternative detection methods,the uniform colorimetric detection method using nano gold particles utilizes the high extinction technology of nano gold particles and the strong distance-dependent optical properties,which can greatly increase the convenience of detection.In this paper,DNA technology is used to construct a DNA calculation model based on nano-gold particles by combining with nano-gold particles,DNA strand replacement technology,bio-enzymatic reaction,DNA self-assembly and other techniques etc.Furthermore,the structure realization of the logic model will be explored in multiple dimensions.Finally,this technology is verified by agarose gel electrophoresis experiments.Firstly,in view of a molecular calculation model for DNA strand displacement based on double nano gold particles,a programmable logic gate structure was designed by using restriction endonuclease as an input signal,specifically identifying DNA cleavage sites through endonuclease,changing the composite structure of gold nanoparticles and DNA,and constructing an output signal.The DNA strand displacement molecular calculation model of the double nano gold particles utilizes the color change characteristics of the nano gold particles caused by particle aggregation or dispersion,and the output signal can be observed with the naked eye,which greatly reduces the complexity and time cost of the detection.Secondly,A molecular calculation model for DNA self-assembly based on single nano gold particles.In this calculation model,the DNA origami technology structure is used to provide a "fulcrum",in addition,considering the "whip chain" and the characteristic of tangent chain which have only seven complementary pairing,the logic gate structure of single nano gold particles has designed,and made test Moreover,the yes,the AND gate and the OR gate are designed further.On account of the gate,the distance control of the structure was completed by changing the "fulcrum",which confirmed the programmability of the calculation model and provided more possibilities for the DNA calculation.
