Objective: to observe the effect of Yao recipe Qingzhen Decoction on Chronic Urticaria Mice with fading evil and seriously damaged qi syndrome and to explore its mechanism,so as to provide reference for the clinical application of Yao recipe Qingzhen Decoction in the treatment of chronic urticaria.Methods: Sixty adult mice were randomly and evenly divided into 6 groups,i.e.the blank group,model group,positive group(diphenhydramine hydrochloride group),and high-,medium-,and low-dose Qingzhen Decoction groups.Except mice in the blank group,all mice in other groups were used to establish Chronic Urticaria Mice model with fading evil and seriously damaged qi syndrome.The mice model was established by subcutaneous injection of 0.3ml/kg reserpine normal saline solution and 2% acetylphenylhydrazine combined with tail vein injection of 0.025% dextran.After one week of adaptive feeding,the blank group and the model group were given normal saline by gavage,and the other groups were given corresponding drugs by gavage,once a day for 10 consecutive days.The experimental observation indicators include:(1)The pruritus of mice in each group was observed after intravenous injection of dextran(mice's scratching incubation period,scratching frequency and itching continuing time)(2)Serum levels of IL-22 and TNF-? were measured by ELISA.(3)the gene expression level of jak1 m RNA and Stat3 m RNA in spleen of mice in each group detected by RT-PCR.(4)the protein expression level of JAK1 and STAT3 detected by Western blot.Results:(1)Except the blank group,different degrees of itching were observed in the other five groups;Compared with the model group,all groups significantly extend the itching scratching incubation period(P<0.01),reduce scratching behavior(P<0.05);shortened the scratching duration(P<0.05).Compared with the positive group,the high-dose group significantly reduced behavior,shortened the scratching duration(P<0.05).There was no significant difference in the itching scratching incubation period between the high-dose group and the positive group(P>0.05)meanwhile,the anti-pruritus effect decreased from the high-dose group to the low-dose group in sequence(P<0.05)(2)Compared with the blank group,the levels of IL-22 and TNF-? were higher in the serum of mice in each group(P<0.05).Compared with the model group,the levels of IL-22 and TNF-? in the serum of mice in other groups were lower(P<0.05).the difference was not statistically significant between the positive group and the medium-dose group(P>0.05).Except that,the other groups were compared in pairs,the difference was statistically significant(P<0.05).(3)Compared with the blank group,the expression of JAK1 m RNA,STAT3 m RNA,JAK1 protein and STAT3 protein in spleen of mice in each group increased significantly(P<0.05).Compared with the model group,the expression of JAK1 m RNA,STAT3 m RNA,JAK1 protein and STAT3 protein was lower in spleen of mice in each group(P<0.05).There were significant differences between the high-,medium-and low-dose groups(P<0.05).the expression levels of JAK1 m RNA,STAT3 m RNA,JAK1 protein and STAT3 protein decreased gradually with the increase of Qingzhen Decoction concentration.Conclusions: Within a certain dosage range:(1)Yao recipe Qingzhen Decoction can improve general conditions of Chronic Urticaria Mice with fading evil and seriously damaged qi syndrome,and reduce their pruritus.(2)The mechanism underlying the therapeutic effects of Yao recipe Qingzhen decoction on Chronic Urticaria Mice with fading evil and seriously damaged qi syndrome may be:(1)reduceing the levels of inflammatory cytokines TNF-? and IL-22 in peripheral blood;(2)inhibiting the expression of JAK1 m RNA,STAT3 m RNA,JAK1 protein and STAT3 protein in the spleen tissue via the JAK/STAT signaling pathway to improve inflammation symptoms.
