Objective:Successfully established a rat model of allergic rhinitis,and observed the effects of Majie Granules combined with Liujunzi Decoction and dried ginger on animal behavioral indicators,HIS,IgE,and IL-4 in serum and nasal mucosa of allergic rhinitis model rats,and To explore its theoretical rules and provide experimental basis for the clinical application of the combined prescription group in the treatment of allergic rhinitis.Methods:Forty-eight healthy SPF-grade SD rats,male and female,were randomly divided into 6 groups of 8 rats each: normal group,model group,combined group,Majie group,Liujunzi group,Xinqin group.In addition to the normal group,the other groups of rats were established with an ovalbumin(OVA)sensitization method to establish an allergic rhinitis model.After the model was successfully established,rats in each group were treated with corresponding drugs.After 14 days of treatment,the rats were anesthetized,aortic blood was drawn,the rats were sacrificed and their nasal mucosa was removed,and HE staining and observation were performed.The serum HIS,IgE,and IL-4were detected.Detection of IL-4 in nasal mucosa by immunohistochemistry.Results:1.Effects on the behavior of AR rats: Compared with the normal group of rats,the AR symptoms of the model group such as nasal scratching,runny nose,and frequent sneezing were prominent and no relief;Compared with the model group,the typical symptoms of AR in the Liujunzi group were not significantly relieved;The above-mentioned allergic rhinitis symptoms in the combination group,hemp mustard group and Xinqin group rats were significantly relieved.2.Effects on the Levels of Related Cytokines in Serum and Nasal Mucosa of AR Model Rats:Compared with normal group rats,the serum IgE content of model group rats was higher than that of normal group rats,the difference was statistically significant(P<0.01);Compared with the model group,the serum IgE content of the rats in the combined group,hemp mustard group,Xinqin group and Liujunzi group was reduced to varying degrees,and the difference was statistically significant(P<0.01);The serum level of HIS in the model group was higher than that in the normal group,the difference was statistically significant(P<0.01);Compared with the model group,the levels of HIS in the serum of rats in the combined group,hemp mustard group and Xinqin group were reducedto varying degrees,and the difference was statistically significant(P<0.01),but still higher than the normal group Medium IL-4 level;The serum level of IL-4 in the model group was higher than that in the normal group,the difference was statistically significant(P<0.01);Compared with the model group,the levels of IL-4 in the serum of rats in the combined group,hemp mustard group and Xinqin group were reduced to varying degrees,and the difference was statistically significant(P<0.01),but still higher than the normal group Medium IL-4 level.The IL-4 content in the nasal mucosa of the model group was higher than that of the normal group;the IL-4 content in the nasal mucosa of rats in the combined group,hemp mustard group and Xinqin group was reduced to varying degrees compared with the model group,but it was still higher than that in the normal group;the IL-4 level of the nasal mucosa of rats in the Liujunzi group decreased slightly,but it was still higher than that of other administration groups.3.Effect on pathological morphology of AR rats:HE staining of the nasal mucosa tissues of rats in each group,the results were observed under light microscope:The nasal mucosa of the normal group was intact,with no obvious abnormalities;in the model group,some nasal mucosa tissues were partially missing and eroded,and the mucosal glands were reduced.Inflammatory cell infiltration and tissue edema were seen.The cilia were obviously shed,showing the characteristics of mucosal remodeling;the nasal mucosa tissue defect of the combined group is not obvious,and the repair condition is good;In the Liujunzi group,the nasal mucosa was still missing and eroded,the submucosal glands were reduced,the inflammatory cells were infiltrated,and a small amount of cilia was still seen;The nasal mucosa of rats in the hemp group still has some defects,the submucosal glands increase,the tissue edema decreases,a small amount of epithelial cells can be seen,and there is no obvious loss of cilia;No obvious defect of nasal mucosa was observed in the Xinqin group,the repair condition was good,and no obvious features of mucosal remodeling were seen.Conclusion: This experimental study shows that the rat model of allergic rhinitis can be successfully prepared using OVA as the allergen;Majie Liujunzi decoction combination formul has an obvious inhibitory effect on the acute phase reaction of allergic rhinitis,alleviating the typical symptoms of AR,and speculates that its mechanism is related to therole of reducing related inflammatory cytokines,and further experimental analysis is needed;Compared with the effect of the Xinqin granules with Majie Liujunzi decoction combination formul on the acute symptoms of AR,the Xinqin granules are superior to the Xinqin granules,but the difference between the two is not obvious,however,according to the clinical practice experience,the long-term curative effect such as prevention of recurrence is ideal,and the next step can be to design relevant experiments for observation and research;It is proved that the use of the combined prescription group based on the theory of Xingyang spleen and lung and spleen homology has advantages in the treatment of allergic rhinitis,so you can also try to apply the combined prescription group method to other allergic diseases,Provide more open ideas and basis for its treatment.
