Study On Lingguishugan Decoction Treatment To Allergic Rhinitis Based On Prescription-syndrome Correlation Theory

ObjectiveThe research intends to explore the relationship of prescription-syndrome correlation between Lingguizhugan Decoction and allergic rhinitis through literature research, clinical research and animal experiments. And then to find the best combination of traditional Chinese medicine dosage of Lingguizhugan Decoction in the treatment of allergic rhinitis by orthogonal design.MethodsThe first part was the arrangements and studies of the documents about the Chinese and western medicine treatment of allergic rhinitis, about prescription-syndrome correlation research. Through the retrieval of the past 5 years (2011-2015) literature, to explore the situation of components of Lingguizhugan Decoction used in the treatment of allergic rhinitis. According to the literature data mining results, these provided the theoretical basis and literature basis for the prescription-syndrome correlation.The second part was clinical investigation and research. Through the information collection of 150 cases diagnosed as allergic rhinitis patients who had phlegm syndrome of TCM, we can explore the relationship between allergic rhinitis and prescription of Lingguishugan decoction. According to the observation of curative effect of 40 patients, it argued that Lingguizhugan Decoction and allergic rhinitis have a certain relationship of prescription-syndrome correlation from the opposite way.The third part was the animal experiments which divided into two animal experiments. In the first experiment,40 SD rats were randomly divided into normal group, model group, Lingguizhugan Decoction group and loratadine group.At the same time, allergic rhinitis model was established. With the corresponding drugs interventions, the therapeutic effect and the prescription-syndrome correlation between Lingguizhugan Decoction and allergic rhinitis was were observed.In the second experiment, according to the drug meter of orthogonal design, the dosages of 4 kinds of component of Lingguizhugan Decoction were divided into two levels and compatibility groups. On this basis, we obtained a quantitative understanding of every factor influencing on Lingguizhugan Decoction to treat allergic rhinitis, such as the best combination of Lingguizhugan Decoction, drug doses and couplet medicines.Results①In the literature of the past 5 years (2011-2015) of TCM in the treatment of allergic rhinitis,378 documents that meet the requirements. Prescriptions for treatment of allergic rhinitis is 776 and 270 kinds of drug were applied. The total drug frequency was 6958 times. The total frequency of four drugs of Lingguizhugan Decoction was 786 times, accounts for about 11.3% of the total frequency, suggests that four drugs of Lingguizhugan Decoction occupy a certain share in the drugs used in the treatment of allergic rhinitis. Fuling dosage was 6-30 g; Guizhi dosage was 3-20 g; Baizhu dosage was 3-30 g;Gancao dosage was 3-15 g. The frequency of appearance of couplet medicines were higher than medication alone. It showed that Lingguizhugan Decoction has a therapeutic effect on allergic rhinitis. ②150 patients of syndrome of phlegm syndrome patients suggested that Lingguizhugan Decoction and allergic rhinitis have a certain relationship of prescription-syndrome correlation. After the use of Lingguizhugan Decoction for 10 days,40 patients had a significant difference in the symptom scores before and after treatment(P<0.05), and the effective rate of the treatment reached 95%. ③In the first animal experiment, allergic rhinitis models were established, containing Model group, Lingguizhugan Decoction group and loratadine group.The symptom scores of the three groups were significantly higher than that of the normal group. After 10 days of treatment, the symptom scores of model group compared with that before drugs used had no significant difference, (P>0.05). The symptom scores of Lingguizhugan Decoction group and loratadine, meanwhile, were distinctly lower than those before administered(P<0.05). Compared with normal group, the number of mast-cell and eosinophil of model group increased significantly. However, Lingguizhugan Decoction group and loratadine group decreased significantly (P<0.05). Compared with normal group, the content of IL-2 of model group was lower and of IL-4 was higher(P<0.05). Compared with model group, the content of IL-2 of Lingguizhugan decoction group and loratadine group were higher, and the content of IL-4 was lower. ④In the test of orthogonal design,8 groups of animal with Lingguizhugan Decoction treatment had a significant difference in the symptom scores before and after treatment (P<0.05). In the end of the experiment, he symptom scores comparison had significant difference among the 8 groups(P<0.05). In the total effectiveness comparison of drugs among the 8 groups, the better combinations of drugs were groupA、G、H. The drugs compatibility proportion of groupA、 H were 4:3:3:2. The combination for Fuling、Guizhi、Baizhu and Zhigancao were 20grams、15grams、15grams and 10grams, respectively, which had the best effect. The relational analysis showed that the levels of Gancao, the mutual effects of Linggui and Lingzhu had significant influence on the curative effects.ConclusionLingguizhugan Decoction and allergic rhinitis have a certain relationship of prescription-syndrome correlation. Allergic rhinitis can be regarded as a kind of phlegm syndrome of nose, Lingguizhugan decoction is effective in treating allergic rhinitis. For the treatment of allergic rhinitis, the best compatibility proportion is 4:3:3:2. And the specific dosages for Fuling, Guizhi, Baizhu and Zhigancao are 20grams,15grams,15grams and 10grams, respectively. The levels of Gancao, the mutual effects of Linggui and Lingzhu have significant influence on the curative effects.
