Objects: To investigate the effect of Xiao'Er Kaiwei zengshi mixture on the expression of Ghrelin,VIP and their receptors in gastric antrum and jejunum of anorexic infantile rats.Methods: 60 SPF grade young SD rats,half male and half female,including 12 blank control group,were fed with conventional feed.Forty-eight mice in the model group were fed special diet by the method of etiology simulation to establish the anorexia model.After successful modeling,the modeling group was randomly divided into four groups,namely model control group,positive control group(jiangzhong jianwei xiaoshi tablet group),Xiao'Er Kaiwei zengshi mixture low dose group(referred to as low dose group),Xiao'Er Kaiwei zengshi mixture high dose group(referred to as high dose group),each group of 12.After 6 weeks of treatment with Xiao'Er Kaiwei zengshi mixture,the children were anesthetized with 10%chloral hydrate and sacrificed by dislocation,and samples were collected.Gastric antrum and jejunum tissues were taken from each rat,and some tissues were routinely fixed with formaldehyde.Histopathological changes were observed and scored under the microscope after HE staining.Part of the organization put in liquid nitrogen,-80 ?,with fluorescence quantitative PCR method and WesternBlot method in rats was detected in the gastric antrum and jejunum tissues Ghrelin,VIP and its receptor mRNA expression and protein expression changes.Results: 1.Pathological sections showed that,compared with the blank control group,the gastric antrum tissue in the model control group had severe mucosal injury,a large number of inflammatory cells infiltration and disordered arrangement of smooth muscle cells.After treatment,inflammatory cells decreased in the positive drug group and the high-dose Xiao'Er Kaiwei zengshi mixture group,and the mucosal structure gradually became complete.For jejunal tissues,compared with the blank control group,the villi in the model control group became shorter and wider,with sparse villi,obvious inflammatory cell infiltration,and a large number of inflammatory cells in the mucosal muscular layer.After treatment,the intestinal villi of the positive drug group and the high-dose Xiao'Er Kaiwei zengshi mixture group increased significantly,widened,the depth of crypt increased,and the inflammatory cells decreased significantly.2.Compared with the blank control group,the mRNA expressions of Ghrelin and its receptor GHSR,VIP and its receptor VPAC2 in the gastric antrum and jejunum tissues of the model control group were significantly down-regulated(P < 0.05),and their protein expressions were significantly decreased(P < 0.05).The mRNA expression of Ghrelin and its receptor GHSR,VIP and its receptor VPAC2 in the gastric antrum and jejunum tissues of the positive control group and the high-dose group were significantly up-regulated(P < 0.05)and the protein expression was significantly increased(P < 0.05)after the treatment with the Xiao'Er Kaiwei zengshi mixture compared with the model control group.Conclusions: 1.Xiao'Er Kaiwei zengshi mixture can reduce the degree of mucosal inflammation and improve the mucosal injury in the gastric sinus tissue of young anorexia model rats.2.Xiao'Er Kaiwei zengshi mixture can improve the length,width,and crypto depth of mucosal villi in jejunum tissue of anorexic model rats,and promote the repair of their villi.3.The expression of Ghrelin and its receptor GHSR,VIP and its receptor VPAC2 mRNA and protein in gastric antrum and jejunum tissues could be up-regulated by Xiao'Er Kaiwei zengshi mixture.Its up-regulation effect on the mRNA and protein expression of Ghrelin and its receptor GHSR,VIP and its receptor VPAC2 may be one of the mechanisms of Xiao'Er Kaiwei zengshi mixture in the treatment of anorexia in children.
