Objiective:This study aims to use the Allergic Rhinitis(AR)model of SD rats as a research object to explore the possible mechanism of the effect of the Sheti zhiqiu formula on the qi deficiency type of AR.The clinical application of the treatment of pulmonary qi deficiency type nasal sputum provides a theoretical basis.Methods:1.40 SPF-grade SD rats were randomly divided into four groups:A,B,C and D,10 in group A(normal group),10 in group B(blank control group),10 in group C(AR model group),and 10 in group D(AR model+traditional Chinese medicine).2.Duplication of AR model of lung-qi deficiency:first duplicate the AR model of lung-qi deficiency syndrome,and then duplicate the AR model of lung-qi deficiency by sensitization.According to Chen Xiaoye's improved smoking method,the model of lung-qi deficiency was duplicated in groups C and D.After 28 days,the behavior of rats with lung-qi deficiency was observed.AR model replication was divided into three stages:systemic sensitization,stimulation sensitization and maintenance sensitization.OVA+A1(OH)~3 combined sensitization was used to sensitize rats by intraperitoneal injection and OVA nasal drip sensitization.After completion,the scores of behavioral symptoms were evaluated.Finally,the samples were collected and the pathological sections were observed by immunohistochemical staining.The expression of TLR-5 protein in rats was detected by parallel optical density,and the concentration of IL-5 in serum was detected by ELISE.Results:The symptomatic behavioral scores of the lung-qi deficiency type AR rats after modeling were all above 5 points.HE staining of nasal mucosa in rats with AR model showed overlap and disorder of mucosal epithelial cells,and cell infiltration such as eosinophils was observed in the cells.ELISE test:serum IL-5 levels in group D(AR model+Chinese medicine group)were lower than those in group C(AR model group).Immunohistochemical staining:TLR-5 was expressed in the nasal septal mucosa of each group,but the expression of TLR-5 protein in the nasal mucosa of group D(AR model+Chinese medicine group)was significantly lower than that of group C(AR model group).Conclusion:1.Sheti zhiqiu formula is effective to improve the systemic and nasal symptoms of lung-deficiency AR rats.2.Sheti zhiqiu formula can reduce the expression of TLR-5 protein in the nasal mucosa of AR rat model and serum IL-5 concentration.3.The mechanism of action of Sheti zhiqiu formula in the treatment of lung qi deficiency type AR may play a role by affecting the expression of TLR-5 protein and then regulating the TH1/Th2 balance shift.
