Objective:1.To discuss the influence of Sheti zhiqiu formula on expression of TLR2 and TLR4 in nasal mucosa and levels of IL-4 and IFN-?in serum of lung-kidney qi deficiency allergic rhinitis patients.2.To establish the model of allergic rhinitis in rats.To observe the influence of LPS on allergic rhinitis rats model.To discuss the influence of Sheti zhiqiu formula on expression of TLR4 in nasal mucosa and levels of IL-4 and IFN-?in serum of allergic rhinitis rats model.Methods:1.Lung-kidney qi deficiency allergic rhinitis patients were chosen as the research objects.The subjects were randomly divided into two groups:the experimental group and the placebo group,and 16 patients in the experimental group and 16 patients in the placebo group.The experimental group were treated with Sheti zhiqiu formula,and the control group were treated with placebo.The course of treatment was 28 days.The expression of TLR2 and TLR4 in nasal mucosa of lung-kidney qi deficiency allergic rhinitis patients were determined by Western Blot and immunohistochemical assay before and after treatment.The levels of IL-4 and IFN-?in serum of lung-kidney qi deficiency allergic rhinitis patients were determined by ELISE before and after treatment.The curative effect of Sheti zhiqiu formula on lung-kidney qi deficiency allergic rhinitis patients were scaled by VAS and RQLQ before and after treatment.2.The 60 clean SD rats were chosen as research objects,and randomly divided into 6 groups.The group A(normal group),group B(blank control group),group C group(AR model),group D(AR model+LPS),group E(AR model+traditional Chinese medicine),group F(AR model+LPS+traditional Chinese medicine)all had 10 rats.Model of allergic rhinitis in rats were established by intraperitoneal injection and nasal topic delivery of ovalbumin(OVA).Abdominal cavity injected with OVA+Al(OH)~3 the next day 1 time(7 times in all)as basis of sensitization.From the 15th day of the experiment,nasal topic delivery with OVA once a day,seven days in a row.AR rats model symptoms integral evaluation were test in the 22th day.From the 22th day of the experiment,nasal topic delivery with low level OVA the next day 1 time,nine days in a row.The group A did not exert any intervention.The group B were intraperitoneal injection and nasal topic delivery with normal saline instead of OVA.The group C were established as standard allergic rhinitis rats model.The group D were established as standard allergic rhinitis rats model and from the22th day of the experiment nasal topic delivery with LPS once a day,nine days in a row.The group E were established as standard allergic rhinitis rats model and from the 22th day of the experiment treated with Sheti zhiqiu formula by intragastric administration once a day,nine days in a row.The group F were established as standard allergic rhinitis rats model and from the 22th day of the experiment nasal topic delivery with LPS and treated with Sheti zhiqiu formula by intragastric administration once a day,nine days in a row.The expression of TLR4 in nasal mucosa of rats model were determined by Western Blot and immumohistochemical staining.The levels of IL-4 and IFN-?in serum of rats model were determined by ELISE.Results:1.The scores of VAS and RQLQ of the experimental group after treatment were lower than that before treatment.The scores of VAS and RQLQ of the placebo group after treatment were lower than that before treatment.There was no significant difference in VAS and RQLQ scores between the experimental group and the placebo group before treatment.There was no significant difference in VAS and RQLQ scores between the experimental group and the placebo group after treatment.The levels of IL-4 in serum of the experimental group after treatment were lower than that in serum of the experimental group before treatment.The levels of IL-4 in serum of the experimental group after treatment were lower than that in serum of the placebo group after treatment.There was no significant difference of the levels of IFN-?in serum between the experimental group and the placebo group before and after treatment.Both TLR2 and TLR4 were expressed in the nasal mucosa of the experimental and placebo groups before and after treatment.The level of TLR2 in the nasal mucosa of the placebo group after treatment were higher than that in the nasal mucosa of the placebo group before treatment.The level of TLR2 in the nasal mucosa of the placebo group after treatment were higher than that in the nasal mucosa of the experimental group after treatment.The level of TLR4 in the nasal mucosa of the placebo group after treatment were higher than that in the nasal mucosa of the placebo group before treatment.2.The combine behavioral scores of allergic rhinitis rats model were all higher than 5 point.The eosinophil infiltration were observed in nasal mucosa of rats model by HE stain.The neutrophile granulocyte infiltration were observed in nasal mucosa of rats model by HE stain after dropped with LPS.The levels of IL-4 in serum of the group E were lower than that in the group C.The levels of IL-4 in serum of the group F were lower than that in the group D.There was no significant difference in serum IFN-?in each groups.TLR4 was expressed in the nasal mucosa of all groups.There was no significant difference in TLR4 expression levels in nasal mucosa among different groups.Conclusion:1.The levels of TLR2 and TLR4 in the nasal mucosa of lung-kidney qi deficiency allergic rhinitis patients would elevate in the course of natural course.2.The levels of TLR2 and TLR4 in the nasal mucosa of lung-kidney qi deficiency allergic rhinitis patients could be decreased by treating with Sheti zhiqiu formula.3.The levels of IL-4 in the serum of lung-kidney qi deficiency allergic rhinitis patients could be decreased by treating with Sheti zhiqiu formula.4.Model of AR in rats had been successfully established.5.The levels of IL-4 in the serum of AR rats model could be decreased by treating with Sheti zhiqiu formula.6.The levels of IL-4 in the serum of AR rats model dropped with LPS could be decreased by treating with Sheti zhiqiu formula.
