Study Of Hepatoprotective Activity Of Qingfei Xiegan Decoction

Qingfei Xiegan Decoction is one of the traditional medicinal prescription to treat Taeumin's disease in traditional Korean medicine.It has been commonly used for the treatment of stroke,arthritis,diabetes and obesity.It is composed of Puerariae Radix,Scutellariae Radix,Angelicae Tenuissimae Radix,Raphani Semen,Platycodi Radix,Cimicifugae Rhizoma,Angelicae Dahuricae Radix,Rhei Rhizoma.The crude drugs of Qingfei Xiegan Decoction was separated two part,one extracted in water and other extracted in ethanol.Both two extracts were separated with petroleum ether,ethyl acetate,w-butanol.In this study,we conducted a vivo pre-experiement to find the extract which has best liver protection activity between water extract,ethanol extract,and othe layer of extract.The result showed the water extracted ethyl acetate layer of and water extract ethyl w-butanol has sig1ificant liver protection activity.The two layers were concentrated and subjected to various chromatographic methods.15 compounds were isolated from water extracted ethyl acetate layer and 6 compounds were isolated from water extracted n-butanol layer of.Their structures were identified as chrysophanol(1),ferulic acid(2),emodin(3),dibutyl phthalate(4),cinnamic acid(5),benzoic acid(6),psoralen(7),bergapten(8),wogonin(9),baicalein(10),daidzein(11),oxypeucedanin hydrate(12),genistein(13),aloe-emodin-3-(hydroxyI-methyl)-O-?-D-glucopyranoside(14),calycosin(15),sissotrin(16),baicalin(17),scutellarin(18),physcion-8-O-?-D-glucopyranoside(19),wogonoside(20),puerarin(21).In vivo experiment,we separated 208 mice to 26 groups,among normal,model and two extracted groups and each layer extract.The Qingfei Xiegan Decoction was given to mice for intragastric administration for 10 days.Then we used the CCI4 induced acute liver injury model in mice.The AST,ALT were measured from serum and SOD,MDA were measured from liver homogenate.It can be seen the water extract showed better protection activity than ethanol extract.The n-butanol layer and thyl acetate layer show better activity than petroleum ether layer under the same extraction conditions.
