Objective:The aim of this study was to examine the therapeutic effect and the mechanism of Qingfeixiegan Decoction on insulin-resistant type 2 diabetic mice.Method:Kun Ming mice were fed with high-fat and high-glucose diet plus streptozotocin to induce insulin-resistant type 2 diabetic mice.Sixty healthy male Kunming rats were fed in the animal feeding room of Yanbian University.All animals had free access to food and water.After acclimatization for one week,mice were randomly divide into six groups.Normal group(CON)fed with standard diet,other five groups fed with high-fat and high-glucose diet including model group(MOD),metformin group(MET,200 mg/kg),Qingfei Xiegan Decoction low-dose group(QXL,200 mg/kg),Qingfei Xiegan Decoction middle dose group(QXM,400 mg/kg),Qingfei Xiegan Decoction high-dose group(QXH,600 mg/kg).Then,the five groups injected with streptozotocin(60 mg/kg)after four weeks.The fasting blood glucose(FBG)and body weight were measured when injected for 72 hours.The mice which fasting blood glucose greater than 11.1 mmol/L were treated with Qingfei Xiegan Decoction and metformin for 28 days,whereas mice in the control and model group were treated with the same volume of physiological saline.Mice with 12 h fasting were euthanized and blood,liver,adipose tissue was collected.The serum biomarkers were measured including FBG,riglyceride(TG),total cholesterol(TC),low density lipoprotein(LDL-C),high density lipoprotein(HDL-C).And the content of Insulin resistance(IR),adiponectin in serum and tumor necrosis factor-a(TNF-a)in serum,liver and adipose tissue were quantified by enzyme-linked immunosorbent assay(ELISA)kits.Result:The results indicated that compared with the normal group,the concentrations of TG,TC,LDL-C,FBG in model group were increased to,respectively.HDL-c was decreased.the level of IR and ADPN in the model group were improved,declined.However,compare to MOD group,the levels of FPF,TG,TC,LDL-C,HDL-C,IR and ADPN in the serum were all reversed in different degrees in QXL,QLM,QLH and MET groups.Moreover,the results of ELISA kit showed that the expression of TNF-a in serum,liver and adipose tissue in model group were increased,respectively,compared with normal group.Thus,Qingfeixiegan Decoction exerted the anti-inflammatory effects of decreasing expression of TNF-a in serum,liver and adipose tissue.Conclusion:Qingfeixiegan Decoction can down-regulate the expression of pro-inflammatory factor TNF-a,up-regulate the expression of ADPN,decrease blood glucose,attenuate hyperlipidemiaand,improve insulin resistance.Our results indicated that Qingfeixiegan Decoction might serve as a potential functional ingredient against diabetes and related inflammation.
