Objective: To study the effect of gauze absorbable on anti-adhesion after cecum and uterine injury in rabbits and rats,the effect of wound healing on the extraction of Bama minipigs after tooth extraction,and to observe the in vitro cytotoxicity of absorbable gauze in order to evaluate its application and safety.Methods: 1.The anti-adherent gauze was leached in a DMEM medium containing 10% fetal bovine serum at a rate of 1.5 cm2/ml,and the extract was used as a test group.High-density polyethylene was used as a negative control,and high-density polyethylene leaching was performed at a rate of 0.2 g/ml serum-containing DMEM medium.The above extracts were immersed in 37 soils at 2°C for 24 hours.A 10% DMSO solution was used as a positive control group,and a blank control group was directly added with 10% fetal bovine serum in DMEM medium.The cytotoxicity of L-929 mouse fibroblasts was measured by MTT assay2.Twenty-four healthy and mature rabbits and 40 SD rats,one female,were not pregnant.Preoperative fasting for 12 hours,can not help but water,using 10% chloral hydrate injection(4mL/kg)anesthesia.Routinely sterilize thedrape,open the abdominal cavity,scrape the cecum and uterus repeatedly with a curette,and cause damage to the serosa layer.The blank control group uses 2mL of saline to smear the wound.The experimental group completely covers the entire wound with absorbable gauze and completes the uterus after completion.With the cecum,close the abdomen.Penicillin and streptomycin were intraperitoneally injected for 3 days,once daily.Observe the absorption and adhesion of absorbable gauze on 3d,7d,and 14 d after surgery,and grade and score the adhesion.3.3 Bama miniature pigs were fed for one week,fasted for 12 hours before surgery,and allowed to drink freely.Anesthesia was performed with ether inhalation and 20% urethane intravenous anesthesia before surgery.The corneal reflex disappeared completely and the animal entered anesthesia.After anesthesia,the oral cavity of the miniature pig was rinsed with 1:5000 potassium permanganate solution and the oral cavity was disinfected twice with iodophor and 75% alcohol.The left and right upper first and third premolars,the left and right lower first premolars were removed and there were 18 teeth in total.Thoroughly clean the wound after tooth extraction and use an ophthalmic scissors to cut the absorbable gauze or the benacel? dental dressing to a suitable size,filling approximately 80% of the socket.The blank group used sterile cotton balls directly.After surgery,observe and record local bleeding and swelling.At 12 h,24h,48 h,3d,7d,and 14 d after operation,the absorption of absorbable gauze,wound healing,and presence of dry trough were observed.Three small pigs were sacrificed 3 days after surgery(Pig No.2),7d(Pig No.1)and 14d(Pig No.3),and the alveolar tissues were fixed with 10% formaldehyde,paraffin-embedded,and routine HE staining.The pathological changes were observed under light microscopeResults: 1.The results of cytotoxicity showed that the positive control group had grade 3 toxicity,and the negative control group had grade 0 toxicity.The experimental samples had different concentrations of extracts and the toxicity was grade 1.Different concentrations of the experimental group and the positive control group were significantly different(P <0.01),different concentrations of the experimental group compared with the negative control group were no significant differences(P> 0.01)2.Rabbit house and cecal adhesions: The incidence of uterine adhesions was: blank group(40.0%),sample A group(75.0%),sample B group(75.0%),sample C group(50.3%);incidence of cecal adhesions They were: blank group(40.0%),sample A group(75.0%),sample B group(75.0%),and sample C group(33.3%).Compared with the blank control group,there was no significant difference in the cecum and uterine adhesion scores between samples A,B,and C(P>0.05).Adhesion of uterus and cecum in rats: The incidence of uterine adhesions was: blank group(44.4%),sample A group(33.3%),sample B group(55.5%),sample C group(50.0%),between each group The adhesion score was not statistically significant(P>0.05);the incidence of caecal adhesions was: blank group(88.9%),sample A group(88.9%),sample B group(55.5%),sample C group(30.0%)and Compared with the blank control group,there was a significant difference in the adhesion score of sample group C(P<0.05),and there was no significant difference between sample groups A and B(P>0.05).3.There was a small amount of bleeding and gingival swelling after tooth extraction in each group.There were dark brown gel masses in the alveolar fossa at 12 h after surgery,and there were 3 cases after 24 h.After 48 h,there were no teeth in the alveolar cavities.See gel,purulent secretions and so on.After 3 days,the wounds in all groups converged,and the gums were slightly swollen and no foreign body was found.On the 7th and 14 th days after operation,the absorbable gauze group,the positive control group,and the blank group healed well,the gums were ruddy and shiny,and there were no complications such as swelling and dry sockets.The results of pathological examination showed that after 3 days,tissue necrosis and bacterial infection were found in all groups.Foreign bodies were also observed in the experimental group and the control group.On the 7th day after surgery,chronic inflammation of the gingiva was associated with fibrosis;fibrosis was evident on the 14 th day after operation.No necrosis,no granulationConclusions: 1.Absorbable gauze has no cytotoxicity2.The incidence of adhesions after cecal lesions in rats is high.Sample C absorbable gauze can prevent adhesions after cecal injury in rats;samples A and B have no obvious anti-adhesion effect after injury to the uterus and cecum.3.Although absorbable gauze and benacel? dental dressings had some gel residue after tooth extraction,there was no significant difference in healing time and histopathology compared with the blank control group,indicating that absorbable gauze could heal wounds after tooth extraction from Bama miniature pigs.No significant effect.
