Objective: To determine genetic polymorphisms of 22 Y-SNP Loci in the non-recombining region(NRY)of 1171 unrelated male individuals from 17 inhabited minorities(Miao,Buyi,Tujia,Dong,Yi,Gelao,Shui,Hui,Bai,Zhuang,Yao,Maonan,Mulao,Mongolia,Qiang,Manchu,She)and Han population in Guizhou.To obtain genetic polymorphism information and data on Y chromosomes of 17 inhabited minorities and Han population in Guizhou.Calculating the genetic distance between groups and building phylogenetic tree among ethnic groups,exploring the genetic relationship between 17 inhabited minorities and Han population and other ethnic groups in Guizhou from the perspective of patrilineal genetics,Providing evidence for molecular evolution for the study of ethnic origin.Methods: A total of 21 Y-SNP loci in 1117 cases of 17 ethnic minorities and Han males in Guizhou were detected by SNap Shot method.The amplified products were analyzed by capillary electrophoresis on ABI 3130 genetic analyzer.ABI 3130 Genetic Analyzer Data Collection Software v3.0 were collected data and obtained 21 Y-SNP typing results.The allele frequency and haplotype frequency were calculated by direct counting method.Haplotype diversity(HD)and Genetic Diversity(GD)were computed by formula:HD or GD= n(1-?Pi2)/(n-1)(Pi is the haplotype frequency or allele frequency,n is the number of samples).The genetic distance were used to calculate using Arlequin3.0 software.Neighbour Joining tree and cluster analysis were established using Mega7.0 software.Results: Among the 21 Y-SNP loci,M407 had a mutation frequency is 0 in 18 ethnic groups,and M175,M214,M9,and M89 had a mutation frequency of 1 in the aquarium population,and there was no polymorphism in the above locus(GD=0.0000),M324 and M122 were polymorphic in 18 ethnic male samples.The M48 locus was polymorphic only in the Dong(GD=0.0298)and Yi(GD=0.0334),and the 47 Z locus was polymorphic only in the Hui(GD=0.0364)and Mongolian(GD=0.0294).The SRY465 site was polymorphic only in the gelo(GD=0.0294)and the Qiang(GD=0.0444)Of the 21 Y-SNP tests in 18 ethnic groups,75 haplotypes were detected in 1171 samples,of which 29 were unique haplotype(UH),H1(125),H2(100),H9(144)and H12(230)are the most important haplotypes.The Fst genetic distance matrix showed that the genetic distance of 18 ethnic groups in Guizhou is between 0.0154 and 0.6573,The smallest genetic distance between Han family and Gelao family is 0.0154;The largest genetic distance between the She population and the Maonan population is 0.6573;Guizhou The genetic distance between 18 ethnic groups and other ethnic groups in China shows that the genetic distance between the 37 ethnic groups is between 0.0008 and 0.658,The smallest genetic distance between Dong family and southern Han family is 0.0008;The largest genetic distance between the She population and the Maonan population is 0.658;the genetic distance between the Guizhou group and the six Asia populations shows that the smallest genetic distance between the Guizhou population and the Philippine population is 0.0611Conclusion: This study obtained the population genetic basis data of 21 Y-SNPs from 17 ethnic minorities and Han nationalities in Guizhou,enriching the genetic information resources of Chinese ethnic minorities.The18 ethnic groups in Guizhou are consistent with linguistic classification and historical sources;.The seven ethnic groups of the Zhuangyu language group contain the Baiyue group characteristic haplogroup(O1a-M119),which confirms the relationship between the ZhuangDonglanguage group and the Baiyue ethnic group.The paternal lineage of the Miao,She,Yao,Qiang,Shui,Maonan and Mulao ethnic groups has a single genetic structure..The genetic structure of the Manchu and Mongolian communities in the paternal line has been gene exchanged with the southern people,and some northern features have been retained.
