ObjectiveTo observe the effect of medicated-catgut embedding at“Ch angqiang"(GV1)on mechanical pain threshold,β-endorphin in hy pothalamus,plasma β-endorphin and nitric oxide content in anus incision pain rats.To explore the mechanism of analgesic effect of medicated-catgut embedding at "Changqiang"(GV1)on anus in cision pain model rats,and to provide scientific basis for the clini cal application.Methods50 SD rats,10 as control group(group A),only after 40 only build mould were randomly divided into model group(group B),G V1-embedding group(group C),medicated-catgut embedding gro up(group D),sham acupoint embedding group(group E),10 in ea ch group.Group A and B are not treated;group C(group of incisio nal pain model with medicated embedding at Changqiang);group D(group of embedding therapy at Changqiang);groupE(group of in cisional pain model with medicated embedding at sham acupoint).and measure rats’ primary pain sensitive and secondary pain sen sitive by Von Frey at preoperative and 4,8,12,24h after operation.The content of beta-EP in the hypothalamus was detected by im munohistochemistry(SP),and the content of beta-EP and NO in peripheral blood was detected by enzyme linked immunosorbent a ssay.ResultsCompared with the control group,the model group,GV1-emb edding group,medicated-catgut embedding group,sham acupoint embedding group,The MWT in the operation area and 1.5 cm awa y from the operation are-a was significantly decreased after 4,8,12,24h(P<0.05),the content of β-EP in hypothalamus was sign ificantly increased at Postoperative 24 hours(P<0.05),the plas ma β-EP content was significantly increased(P<0.05),the plas ma no content was significantly increased(P<0.05).compared wi th the model group,GV1-embedding group,medicated-catgut emb edding group’s MWT in the operation area was significantly increa sed after 8,12,24h(P<0.05).The MWTat1.5 cm away from the o peration area was significantly increased after12,24h(P<0.05),t he content of β-EP in the hypothalamus increased significantly(P<0.05),the plasma β-EP content was significantly increased(P<0.05)and the content of plasma no decreased significantly(P<0.05),compared with the medicated-catgut embedding group.the GV1-embedding group’s MWT in the operation area was sign ificantly increased after 12,24h(P<0.05),The MWTat1.5 cm aw ay from the operation area was significantly increased after 24h(P<0.05).the content of β-EP in the hypothalamus increased si gnificantly(P<0.05),the plasma β-EP content was significantly increased(P<0.05)and the content of plasma no decreased sign ificantly(P<0.05)There was no significant difference in the 24h hypothalamic beta EP,plasma beta EP and NO level between the model group and sham acupoint embedding group.(P>0.05).Conclusionsmedicated-catgut embedding at "Changqiang"(GV1)could sig nificantly increase the mechanical pain threshold,increase the co ntent of β-EP in hypothalamus and plasma,and decrease the con tent of no in plasma.
