| Objective:Observing acusector for ovaries of mammary gland hyperplasi a model rats impact of Breast shape(nipple diameter,altitude),breast tis sue pathology and hormone(GnRH?PRL),explore the curative mechanism of tr eating MGH further.Methods:Choosing 54 female SD rats,healthy,unfertilized maturation.Fe eding 1 week for environmental adaption.Dividing 54 rats into two groups randomly,15 rats as blank group(Group A),39 rats as modeling group.Rep licating model for modeling group by MGH:Injecting estradiol benzoate of0.5 mg/kg in the lateral thigh muscle,1/d,for 20 d.Then injecting proges terone at the same place for 5mg/kg,1/d,5d.Blank group were injected amo unt 0.9% saline,1/d,for 25 total days.Selecting three rats from each gro up randomly next day and cutting the second line of breast tissue then p athology observation to confirm the successful modeling.Dividing the res t 36 rats of modeling group into model group(group B),ovarian group(grou p C),ovarian electric acupuncture group(group D)based on random number t able method,12 rats each group.All rats fasting for solids and liquids f or one day.As for all rats in Group C and Group D,Remove their ovaries,(In operation,1 rat in Group D died).Injecting intramuscular penicillin0.075g/kg for each rat for 3 consecutive days after the surgery,having a electric acupuncture treatment for Group D next day.The selected points in Group A are Tianzong,Ganshu,Zusanli,both bilaterally The selected po ints in Group B are Wuyi,Hegu(both bilaterally),Danzhong.Two groups of a cupoints used interchangeably,1 set per day.Both side Tianzong and Gansh u,both side Wuyi and Hegu connect two groups of electrodes,respectively.Tianzong and Wuyi connect the positive,Ganshu and Hegu connect the catho de.Through to G6805-type Cupping therapy instrument 2 hz,dilatational wa ve,treatment for 20 minutes at a time,treatment 1times,5 times for a period of treatment.The rest 2d,on to the next period of treatment,a total of four courses of treatment.Groups of rats daily in addition to seize,r egular disinfection.Don't do any other processing.After the treatment,fa sting and ban water for Group A,B,C and D.for 1 day.Then measure each ra ts nipple height and diameter,and cut the full breasts,paraffin embeddin g,sectioning.Then observe the changes of breast tissue with light micros cope.Abdominal aortic blood again,centrifugal in serum low temperature p reservation.waiting to detect the contents of GnRH,PRL.Results:1.The height and diameter of thenipple:(1)The height of the nipple:Compared with the model group,the heigh t of the nipple decreased,the difference was significant(P<0.01).Compar ed with ovariectomized group,the papillary height of ovariectomized rats was lower than that of ovariectomized group,the difference was existed(P<0.05).(2)The diameter of nipple:Compared with the model group,the diameter of the nipples in the ovariectomized group was smaller,the difference w as very obvious(P<0.01);Compared with the ovariectomized group,the diam eter of the nipples of the ovariectomized rats was smaller,the differenc e was existed(P<0.05).2.Changes of mammary histology under light microscope:In the blank group,the ductal epithelium of mammary duct was arranged in an orderly manner,there was no dilatation of the lumen of the cathet er and the alveolus;The mammary glands of model group were diffusely pro liferated,acini and lobules increased significantly,fat and connective t issue decreased significantly,the arrangement of ductal epithelial cells is disorder,with epithelial hyperplasia clearly,the acinar cavity and the lumen of the catheter were dilated markedly,intraluminal epithelial cells and secretions can be seen in the lumen,prompt model replication s uccessful;Compared with the model group,the mammary gland lobular volume of the ovariectomized group decreased,the epithelial cells of the ducta l hyperplasia improved,and the number of acini decreased,but there are still some acinar cavities and dilated lumen,and the secretions in the ac inar cavity decrease;Compared with the ovariectomized group,the arrangem ent of mammary ductal epithelial cells in the EA group tended to be tidy,the dilatation of the lumen and lumen of the acinus decreased significa ntly,and the number of lobules decreased and the number of acini decreas ed significantly.3.Changes of serum GnRH and PRL levels:(1)GnRH:Compared with the model group,the serum GnRH levels of ovarie ctomized rats were increased,the difference was very significant(P<0.01);Compared with the ovariectomized group,the serum GnRH levels in ovariec tomized rats were significantly lower than those in the OVX group(P<0.01).(2)PRL:The level of PRL in the ovariectomized group was significantly lower than that in the model group(P<0.01);Compared with the ovariectom ized group,the level of PRL decreased,the difference was significant(P<0.01).Conclusion:1.Ovaries removing reduces MGH rats' nipples increased and micrifies diameter increased,which shows the MGH rat breast shape is effected by o varies removing;Electric acupuncture can make the remove ovarian MGH nip ple height and diameter of rats tend to be normal,which explains ovarian path is the method for electric acupuncture treatment MGH.2.Electric acupuncture can obviously improve the disorder of glandula r structure,make MGH rats breast duct neatly,lumen,acinus cavity signifi cantly narrowed,secretion in Lumendecreased significantly.3.Electric acupuncture may reduce high concentration of remove ovaria n MGH rats GnRH and PRL level,which explains the mechanism of electric a cupuncture treatment is fulfilled based on diseasing serum content of Gn RH hypothalamus and pituitary PRL of MGH rats. |
