Purpose:This research was designed to observe the effect of acupuncture at the acupoints“danzhong”(CV 17), “zusanli”(ST 36), and “taichong”(LR 3) on the quantity of estradiol(E2)and progesterone(P) in serum, the expression of estrogen receptor(ER) m RNA and progesterone receptor(PR) m RNA in breast tissue, gonadotropin releasing hormone(Gn RH)m RNA in hypothalamic and gonadotropin releasing hormone receptor(Gn RH-R) m RNA in pituitary gland in rats with hyperplasia of mammary gland(HMG).We did the whole research in order to explore if the abnormal expression of the gonadal hormones and their corresponding receptors of the hypothalamic-pituitary-ovary axis(HPO axis) is one of the pathogenic factors of HMG and clear the curative effect and mechanism of acupuncturing at the acupoints “danzhong”(CV 17), “zusanli”(ST 36), and “taichong”(LR 3) in the treatment of HMG.Materials and methods:40 female and non-pregnant SD rats(SPF grade), were randomly divided into 4 groups(n=10), respectively called the normal group, the model group, the acupuncture group and the taxoxifen group. After duplicating modeling, 4 weeks of treatment began: the rats in the normal group were hold once time daily; the rats in the model group were tied up only on the rabbit set for 15 min daily; the rats in the acupuncture group were tied up on the rabbit set for acupuncturing at “danzhong”(CV 17) point and electro-acupuncturing at the acupoints“zusanli”(ST 36) and “taichong”(LR 3) for 15 min daily; the rats in the taxoxifen group were given taxoxifen(1.8mg/kg) once time daily.After the treatment, the diameter of the second pairs and the third pairs of nipples(left) were measured. And then we took the second pairs and the third pairs of breast, the serum, the hypothalamic and the pituitary gland out of the rats in each group. The second pairs of breast were used for observing pathological changes of the mammary gland by HE staining method; the serum was used for observing the quantity changes of E2 and P by Elisa; the third pairs of breast were used for observing the expression of ER m RNA and PR m RNA by RT-PCR; the hypothalamics were used for observing the expression of Gn RH m RNA by RT-PCR; the pituitary glands were used for observing the expression of Gn RH-R m RNA by RT-PCR.Results:1 The mean value of the nipple diameter in each group:Compared with the normal group, the nipple diameters in the model group were higher obviously(P<0.01); compared with the model group, the nipple diameters in the acupuncture group and the taxoxifen group were lower obviously(P < 0.01); there was no significant difference between the acupuncture group and the tamoxifen group(P>0.05).2 Pathological perfpormance in the breast tissue of rats in each group:Compared with the normal group, in the breast tissue of rats in the model group, the acinis were increased in volume and quantity, and the arrange of the acinis were irregular, the excreta in acinis and mammary ducts were increased, the performance of hyperplasia of mammary gland was obvious; Compared with model group, in the breast tissue of rats in the acupuncture group and the tamoxifen group, the acinis were reduced in volume and quantity,and the arrange of the acinis were in neat rows, the excreta in acinis and mammary ducts were reduced, the degree of hyperplasia of mammary gland was unconspicuous.3 Quantity of E2 and P in serum in each group:Compared with the normal group, the values of the E2 in the model group were higher obviously(P<0.01) and the values of the P were lower obviously(P<0.01); compared with the model group, the values of the E2 in the acupuncture group and the tamoxifen group were lower obviously(P<0.01) and the values of the P were higher obviously(P<0.01); there was no significant difference between the acupuncture group and the tamoxifen group(P>0.05).4 The expression of ER m RNA and PR m RNA in breast tissue, GnRH m RNA in the hypothalamics, Gn RH-R m RNA in the pituitary glands in each group:Compared with the normal group, the expression of ER m RNA, PR m RNA, GnRH m RNA and Gn RH-R m RNA in the model group were higher obviously(P<0.01); compared with the model group, the expression of ER m RNA, PR m RNA, Gn RH m RNA and Gn RH-R m RNA in the acupuncture group and the taxoxifen group were lower obviously(P<0.01); there was no significant difference between the acupuncture group and the tamoxifen group(P>0.05).Conclusion:1 The treatment of acupuncturing at the acupoints “danzhong”(CV 17), “zusanli”(ST 36), and“taichong”(LR 3) can improve the hyperplasic nipples and pathological changes of the breast tissue in the model rats of HMG. So the conclusion that the treatment of acupuncturing at the acupoints “danzhong”(CV 17), “zusanli”(ST 36), and “taichong”(LR 3) has therapeutic effect for HMG is proved.2 The abnormal changes of the gonadal hormones and their corresponding receptors of HPO Axis is closely related to the occurrence of HMG.3 The mechanism of acupuncturing at the acupoints “danzhong”(CV 17), “zusanli”(ST 36),and “taichong”(LR 3) in the treatment of HMG is its regulating role to the expression of the gonadal hormones and their corresponding receptors of HPO Axis.
