Genetic Polymorphism Analysis Of 6 Y Chromosomal STRS In Korean And Han Population In China

Objective:To observe genetic polymorphism of 6 Y-STRs among 200 unrelated Chinese Korean males and 200 unrelated Han males in Chinese by multiplex amplification system,which to obtain the forensic genetic statistical data of the two groups.Aim at establishing Chinese Korean DNA database as well as offering basical data for Individual identification,Paternity identification,Human Genetics and other objects.Methods:Tested 6 Y-STRs of 200 unrelated Chinese Korean and 200 unrelated Han individuals by PCR and detected by polyacrylamide gel electrophoresis with Silver staining.The results were calculated by Excel 2016,SPSS,Arlequin 3.5 and Mega4.1.Results:There were 13?6?4?4?5 alleles detected from Chinese Korean population in single-copy locus DYS449?DYS461?DYS508?DYS531?DYS594,and Gene Diversity were 0.8436?0.6047?0.5662?0.5303?0.3310 while there were 11?6?5?4?6 in Han population and Gene Diversity were 0.8690?0.6344?0.4815?0.5511?0.3850.Muti-copy locus DYF404S1 was observed 26 haplotypes in Chinese Korean and 24 in Han,however,Haplotypes Diversity were 0.9260 and 0.9107,Discrimination Power is 0.9214 and 0.9062 of the two population respectively.In Chinese Korean groups,the total locus of the experiment composed 167 haplotype and its HD,DC,MP,DP were 0.9981,0.8350,6.85 × 10-3,0.9932.Among Han population,the 6 Y-STRs accumulated 181 haplotype and its HD,DC,MP,DP were 0.9989,0.9050,6.05 × 10-3,0.9940.Moerover,compared two groups with eleven differents ethnic around the world for obtaining P-value and genetic distance.Conclusion:Rapidly mutating Y-STRs including in DYS449 and DYF 404S1 were highly diverse in Individual identification and Paternity identification in Chiense Korean Population as well as in Han,while DYS461,DYS 508,DYS531 were observed the same phenomenon,but other loci DYS594 were not.The data of 6 Y-STRs in two populations in the two population can provide basical data for Chinese Korean database and in forensic Individual identification,Paternity identification,Human Genetics or other objects.
