Genetic Polymorphism Of Six Y-STR Loci From Chinese Korean And Han Males

Objective:to study the distribution of genetic polymorphism with six Y chromosome STR loci DYS19, DYS388, DYS390, DYS391, DYS392, DYS393from Chinese Korean and Han males, for providing reference data in Forensic medicine Individual identification, Paternity identification, anthropology and other related fields.Methods:(1) Anticoagulation specimens were collected from200unrelated Chinese Korean males and200Chinese unrelated Chinese Han males. The DNA were extracted with blood genomic prep mini kit.(2) DYS19, DYS388, DYS390, DYS391, DYS392, DYS393loci were tested by amplifying the techniques of PCR.5%polyacrylamide gel electrophoresis and silver-staining to200Chinese Korean males and200Chinese Han males. The data were analyzed with Arlequin2000.(3)The alleles of each Y-STR loci were sequenced and the allele ladders were prepared. According to the recommendation of International Society of Forensic Genetics (ISFG).Results:Total6,4,5,5,6.5alleles were observed at DYS19, DYS388, DYS390、 DYS391, DYS392, DYS393loci in Chinese Koreans respectively and the calculated gene diversity(GD) were0.6722.0.4848.0.6674,0.2641,0.6704;0.6405,respctively,six Y-STR loci were accumulatived and analyzed, a total of109heliotypes were found, and heliotypes diversity was0.9803;In all6,5,7,5,7,5alleles observed in Chinese Han males and calculated gene diversity(GD) were0.7130,0.5219,0.6971,0.3509,0.7439,0.6564, respectively. Six Y-STR loci were accumulatived and analyzed total133heliotypes were found, and heliotypes diversity was0.9914Conclusion:The data displayed from the experiment. Six Y-STR loci have better genetic polymorphism and stability and highly applied value except genetic polymor- phism of DYS391was lower. The data can be provided in medical jurisprudence individual identification, paternity test and human genetics investigation for Chinese Korean males and Chinese Han males.
